Method for the synthesis of nucleotide derivatives wherein molecules of interest are grafted on the oligonucleotide with the help of a 'click chemistry' reaction between an azide function on the molecule of interest and an alkyne function on the oligonucleotide, or between an alkyne function on the molecule of interest and an azide function on the oligonucleotide. Intermediate molecules, notably alkyne functionalized oligonucleotides, grafted oligonucleotides, azide functionalized oligonucleotides, oligonucleotide micro arrays containing them and the use of those grafted oligonucleotides for biological investigation and for cell targeting.

Method for the synthesis of oligonucleotide derivatives

A method for mixing and dispensing compounds, at least one reactant in a carrier fluid in laminar flow, and a cell for implementing the method. The cell includes a reaction chamber having a reaction surface where the reactant can be fixed, directly or indirectly, at least three fluid inlets/outlets, a fixed volume reservoir in communication with the reaction chamber and outside the reaction chamber via injection orifice (), a fluid loop including a feeding port, an extraction port and a variable volume reservoir adapted to communicate independently with the reaction chamber via each of its inlets/outlets; device for circulating fluids in the reaction chamber and the fluid loop. The method and cell can be used to prepare homogeneous surface films, or to carry out 'target-probe' identification reactions.

The invention relates to a biochip comprising a substrate presenting a reflecting main surface and wherein the main surface presents localized sites that are made sensitive to fluorescence detection by a transparent layer having optical thickness (2k+1)&#x3bb;/4 where k is a positive or zero integer and where &#x3bb; designates a wavelength lying between a wavelength &#x3bb;at which fluorescence is excited and a wavelength &#x3bb;at which fluorescence is emitted. The biochip can be manufactured by depositing a transparent layer of thickness m&#x3bb;'/2 on a substrate and by making studs or wells of thickness (2k+1)&#x3bb;/4 in said layer to form said sensitive sites.
