An apparatus is disclosed which is capable of detecting a selected gas in a liquid sample to a sensitivity in the range of 10 to 10,000 parts per trillion. Such sensitivity is achieved by concentrating the selected gas in a sample without concentrating sources of ionic interference that are also present in the sample. Selective concentration of the gas is achieved by vaporizing the sample, correcting for non-ideal vaporization, selectively condensing the selected gas from the vapor, and detecting the selected gas. Methods for using the apparatus and detecting the selected gas are also disclosed.

Parts per trillion detector

A method and apparatus for the measurement of the total organic carbon (TOC) content of a liquid is shown. The inorganic carbon in the liquid is converted into carbon dioxide and removed from it. At the same time, oxygen is added to the liquid. The liquid is then exposed to ultraviolet radiation and the organic carbon thereby oxidized. This oxidized carbon, in the form of carbon dioxide, is allowed to migrate across a gas-permeable, liquid-impermeable membrane directly into a measurement chamber, where its absorption of radiation is measured to determine its concentration.

Method for reagentless measurement of the total organic carbon content

A method and apparatus are disclosed for determining the concentration of coliform bacteria in a sample. The sample containing the coliform bacteria is cultured in a liquid growth medium. The cultured bacteria produce hydrogen and the hydrogen is vented to a second cell containing a buffer solution in which the hydrogen dissolves. By measuring the potential change in the buffer solution caused by the hydrogen, as a function of time, the initial concentration of bacteria in the sample is determined. Alternatively, the potential change in the buffer solution can be compared with the potential change in the liquid growth medium to verify that the potential change in the liquid growth medium is produced primarily by the hydrogen gas produced by the coliform bacteria.

Method for detecting coliform organisms

A bioluminescent assay for ATP in water borne bacteria is made by adding nitric acid to a water sample with concentrated bacteria to rupture the bacterial cells. The sample is diluted with sterile, deionized water, then mixed with a luciferase-luciferin mixture and the resulting light output of the bioluminescent reaction is measured and correlated with bacteria present. A standard and a blank also are processed so that the light output can be correlated to bacteria in the sample and system 'noise' can be substracted from the readings. A chemiluminescent assay for iron porphyrins in water borne bacteria is made by adding luminol reagent to a water sample with concentrated bacteria and measuring the resulting light output of the chemiluminescent reaction. The light output is correlated with bacteria present. A standard and a blank are also processed so that the light output can be correlated to bacteria in the sample and system 'noise' can be subtracted from the readings. Modifications may be made in the methodology to differentiate between live and dead bacteria. An automatic system automatically performs a biolumenscent ATP assay on a concentrated bacterial sample. Reservoirs are provided for the sample, standard and blank. These are sequentially mixed with nitric acid from a reservoir by using two channels of a peristaltic pump. This acid mixture is then mixed with sterile, dionized water from another reservoir using two additional channels of a peristaltic pump and the resulting mixture is then mixed with a luciferase-luciferin mixture from an additional reservoir by employing two more channels of a peristaltic pump. The resulting solution flows through a photometer which indicates the level of the bioluminescent light reaction.

Rapid, quantitative determination of bacteria in water

Method and automated apparatus are disclosed for determining the time of detection of metabolically produced hydrogen by coliform bacteria cultured in an electroanalytical cell from the time the cell is inoculated with the bacteria. The detection time data provides bacteria concentration values. The apparatus is sequenced and controlled by a digital computer to discharge a spent sample, clean and sterilize the culture cell, provide a bacteria nutrient into the cell, control the temperature of the nutrient, inoculate the nutrient with a bacteria sample, measures the electrical potential difference produced by the cell and measures the time of detection from inoculation.

Method and automated apparatus for detecting coliform organisms

A method and apparatus for removing porphyrins from a fluid sample which are unrelated to the number of bacteria present in the sample and prior to combining the sample with luminol reagent to produce a light reaction. The method involves a pre-incubation of the sample with a dilute concentration of hydrogen peroxide which inactivates the interfering soluble porphyrins. Further, by delaying taking a light measurement for a predetermined time period after combining the hydrogen peroxide-treated water sample with a luminol reagent, the luminescence produced by the reaction of the luminol reagent with ions present in the solution, being short lived, will have died out so that only porphyrins within the bacteria which have been released by rupturing the cells with the sodium hydroxide in the luminol reagent, will be measured. The measurement thus obtained can then be related to the concentration of live and dead bacteria in the fluid sample.
