Particle detection cartridges are provided. Aspects of the particle detection cartridges according to certain embodiments include a sample input, a flow channel and a light channel, where the flow channel and light channel are coupled at a detection region such that only light from the detection region can propagate directly through the light channel to a detector. Systems including the cartridges, as well as methods for detecting particles in a sample with the subject particle detection cartridges/systems, are also described. Kits having one or more cartridges are also provided.

Particle detection cartridges, systems thereof and methods for using the same

Aspects of the present disclosure include methods and systems for assaying a sample for an analyte. Methods according to certain embodiments include illuminating a sample with a slit-shaped beam of light, detecting light transmitted through the sample, determining absorbance of the transmitted light at one or more wavelengths and calculating concentration of the analyte based on the absorbance to assay the sample for the analyte. Systems for practicing the subject methods are also described.

Optical imaging system and methods for using the same

The present disclosure provides methods and systems for assaying a sample. A microfluidic device to perform an assay of a sample (e.g., biological sample) is described having a sample application site, a porous component and a flow channel. The porous component provides for uniform dissolution of a reagent and mixing of the sample and reagent without filtering the sample.

Microfluidic devices, and methods of making and using the same

The present disclosure provides cytometric methods for the detection of rare target cells in a sample. In certain aspects, the methods and compositions may facilitate the detection of rare target cells, such as circulating tumor cells (CTCs), in a biological sample such as blood. Aspects of the methods include contacting the sample with first and second binding members that specifically bind to a marker of the rare target cell, and cytometrically assaying the sample for the presence of cells comprising bound first and second binding members to detect the rare target cell in the sample. Also provided are systems, compositions, and kits for practicing the subject methods.

Methods and compositions for cytometric detection of rare target cells in a sample

The present invention provides a low cost imaged-based system for detecting, measuring and/or counting labeled features of biological samples, particularly blood specimens. In one aspect, the invention includes a system for imaging multiple features of a specimen that includes one or more light sources capable of successively generating illumination beams each having a distinct wavelength band and a plurality of differentially excitable labels capable of labeling a specimen comprising multiple features, such that each different feature is labeled with a different differentially excitable label.

Method, system, and compositions for cell counting and analysis

The present invention provides a low cost imaged-based system for detecting, measuring and/or counting labeled features of biological samples, particularly blood specimens. In one aspect, the invention includes a system for imaging multiple features of a specimen that includes one or more light sources capable of successively generating illumination beams each having a distinct wavelength band and a plurality of differentially excitable labels capable of labeling a specimen comprising multiple features, such that each different feature is labeled with a different differentially excitable label. System of the invention may further include a controller operationally associated with the one or more light sources for successively directing illumination beams onto the specimen so that each of the different differentially excitable labels is successively caused to emit an optical signal within the same wavelength band, an optical system capable of collecting such emitted optical signals and forming successive images corresponding to the labeled features of the specimen on a light-responsive surface to form successive sets of image data thereof, and a disposable cuvette for collection and optical analysis of non-red blood cells.

The invention provides methods and compositions for identifying and counting lymphocytes in a biological sample, such as whole blood, by means of a probe comprising at least one binding compound specific for a T lymphocyte-specific marker, e.g. a CD2 or CD3, and at least one binding compound specific for CD45RA. Lymphocytes within the sample combine with such a probe to form a distinguishable subpopulation based on the amount of probe that specifically binds to their surfaces, thereby permitting such lymphocytes to be detected and enumerated on the basis of the intensity of the signal generated by the probe, and without the need of a separate physical measurement, such as light scatter. With additional probes specific for additional blood cell markers, percentages of lymphocytes, monocytes and granulocytes in a sample may be determined.

Method of analyzing lymphocytes

A method and apparatus for autofocus on a target layer contained within a microplate well is provided. The instrument is capable of optically sensing a reference point on the underside of a microplate. This reference point is then used to focus light onto a target layer within the microplate well, the target layer having a location that is in defined relation to the reference point. The reference point is either a surface of the bottom of the microplate well or is an optically detectable mark on the underside of the microplate. In an alternate embodiment, a light position sensitive detector is used to enable deterministic autofocus for a plurality of wells on a microplate.

A catheter for introducing fluid into the vascular system includes ports in the catheter's sidewall which are obliquely angled with respect to the longitudinal axis of the catheter. Certain pairs of ports are oriented toward each other, and certain other pairs of ports are oriented away from each other. The arrangement of ports causes fluid to exit the ports in streams which are oriented at oblique angles corresponding to the angles of the port. The obliquely angled streams create advantageous circulation paths proximate to the catheter which generally improve efficacy of the injected fluid associated with the medical application. The catheter may be used for a variety of purposes, including to perform thrombolysis.
