Metallo-desferrioxamine complexes and their use in the treatment of bacterial infections

The invention presented herein provides methods and compositions for the prevention and treatment of bacterial infections. The methods are based on the discovery that depletion of bioavailable iron stimulates surface motility in bacteria thus inhibiting the ability of a bacterial population to develop into a biofilm.

Methods of inhibiting and treating bacterial biofilms by metal chelators

The present inventions are directed to a novel bactobolin analog bactobolin D. The present inventions also are directed to a method of producing a composition comprising at least one bactobolin analog using a bacteria strain comprising a bacterial cell comprising the biosynthetic locus of the bactobolin analog in(bta cluster) or a homolog structure (at least 95% sequence identity) thereof, and further isolation and purification of the bactobolin analog. For example, the bacterial strain can be a wild type bacterial strain such as astrain comprising a bta cluster (e.g.(e.g. E264, Bt4, and TXDOH) and(e.g. K96243, 1106a, and 1106b).

Provided are compositions and methods useful for modulating the activity of autoinducer synthase catalysts. A method for identifing modulators of the autoinducer synthesis reaction is also provided. Such modulators are useful for controlling bacterial growth and can be used for therapeutic treatment of bacterial infections particularly in immunocompromised subjects. They are also useful in treating disease states associated with autoinducer synthesis and biofilm development.
