Disclosed are processes for forming compositions comprising small domains of an active agent and a matrix material, and methods of using them. A suspension of an active agent, a matrix material, a first solvent, and a second solvent is formed at a temperature T, heated to a temperature Tand spray dried.

Process for forming active domains dispersed in a matrix

A pharmaceutical composition comprises multiparticulates comprising a melt-congeal core and a solid amorphous dispersion layer of a poorly water soluble drug and polymer. The multiparticulates are suitable for improving bioavailability of poorly water soluble drugs. The melt-congeal cores facilitate application of the solid amorphous dispersion layer, and allow incorporation of additional optional components to the core so as to adjust the release of drug from the multiparticulate.

The process comprises delivering a spray solution comprising an active agent and a matrix material in an organic solvent to a spray-drying apparatus, atomizing the spray solution into droplets within the spray-drying apparatus to remove at least a portion of the organic solvent from the droplets to form a plurality of particles, and collecting the particles. The spray solution may be formed by forming a feed suspension comprising the active agent, the matrix material, and the organic solvent, wherein the feed suspension is at a temperature T, and directing the feed suspension to a heat exchanger, thereby increasing the temperature of the feed suspension to a temperature T, wherein Tis greater than T, and the spray solution is at a pressure greater than the vapor pressure of the organic solvent at T, such that the active agent and matrix material are soluble in the organic solvent at T.

Oral rehydration solutions (ORSs), methods of preparing ORSs and methods of delivering rehydration therapy are disclosed herein. In some embodiments, an ORS includes a non-starch viscosity enhancing polymer and a sodium salt. The ORS may include at least approximately 50 millimoles of dissolved sodium per liter. Pharmaceutical compositions are provided comprising an active agent and a dextran polymer derivative. The compositions include from 0.01 to 99 wt % of an active agent and from 1 to 99.99 wt % of a dextran polymer derivative.
