Electrospun nanofibrils and methods of preparing the same are provided. The electrospun nanofibrils comprise at least one polypeptide. A polypeptide can be dissolved in a solution, and the solution can be electrospun into a nanofibril. The solution can be added to a syringe or syringe pump, and an electric field can be applied to electrospin the at least one polypeptide.

The subject invention pertains to peptide-based materials comprising cross-linked peptides with random amino acid sequences that are soluble in water or ethanol before crosslinking but insoluble in water after crosslinking.

Disclosed herein are polypeptide multilayer films wherein a therapeutic agent is covalently linked to a first layer polypeptide. An advantage of such attachment is that the linked therapeutic agent can be controllably released from the multilayer film into the environment of the film upon the addition of a suitable stimulus. An advantage of the disclosed films and methods is enablement of environmentally-stimulated release under specific conditions.

Disclosed herein is a multilayer film comprising two or more layers of polyelectrolytes, wherein adjacent layers comprise oppositely charged polyelectrolytes. A first layer polyelectrolye comprises a composite polypeptide comprising one or more surface adsorption regions covalently linked to one or more functional regions forming a single polypeptide chain. The surface adsorption regions comprise one or more amino acid sequence motifs consisting of 5 to 15 amino acid residues. The one or more functional regions comprise 3 to about 250 amino acid residues.

Multilayer films, coatings, and microcapsules comprising polypeptides

Disclosed herein are polypeptide multilayer films comprising a hybrid polypeptide comprising a first polypeptide segment and a second segment, the two segments being covalently joined by one or more non-peptidic linkages. The first segment comprises a polypeptide having a magnitude of net charge per residue of greater than or equal to 0.4, and a length of greater than or equal to about 12 amino acid residues. The second segment comprises a polypeptide or another polyelectrolyte.

Disclosed herein are immunogenic compositions comprising a multilayer film comprising two or more layers of polyelectrolytes, wherein adjacent layers comprise oppositely charged polyelectrolytes. A first layer polyelectrolyte comprises an antigenic polypeptide comprising one or more surface adsorption regions covalently linked to one or more antigenic determinant regions, wherein the antigenic polypeptide and the one or more surface adsorption regions have the same polarity. The immunogenic compositions may be employed in methods of eliciting an immune response in a vertebrate organism.

Disclosed herein is a method of making a film, the method comprising depositing a first layer polyelectrolyte on a surface of a substrate to form a first layer; and depositing a second layer polyelectrolyte on the first layer polyelectrolyte to form a second layer. The first layer polyelectrolyte, the second layer polyelectrolyte, or both, is deposited on the substrate in the presence of a polymeric precipitant; and the first layer polyelectrolyte and the second layer polyelectrolyte have net charges of opposite polarity. Also disclosed are methods of improving bioactive molecule retention during fabrication of a polyelectrolyte multilayer film.
