Provided are methods for preparing gelled, solubilized extracellular matrix (ECM) compositions useful as cell growth scaffolds. Also provided are compositions prepared according to the methods as well as uses for the compositions. In one embodiment a device, such as a prosthesis, is provided which comprises an inorganic matrix into which the gelled, solubilized ECM is dispersed to facilitate in-growth of cells into the ECM and thus adaptation and/or attachment of the device to a patient.

Extracellular matrix-derived gels and related methods

The vasculature of a donor lung is perfused with a lung preserving fluid to preserve its structure. At the same time, a decellularization fluid is perfused through the airways, which strips away donor cells. The decellularized region is then seeded with pulmonary cells of the transplant recipient, which regenerate the lung. The pulmonary cells may be derived from stem cells, and the decellularization can be targeted to reduce the quantity of cells required.

Functional recovery of human lungs for transplantation

Disclosed are bioreactor devices, systems and methods. A bioreactor system can include one or more bioreactor modules that can be individually controllable and identifiable. A bioreactor module can be connected to one or more functional modules such as a pump module, a stimulation signal generation module, a motor module, a mechanical transmission module, a gas exchange module, a temperature module, a humidity module and/or a COmodule, among others. The bioreactor and functional modules can include standard or universal connectors to facilitate connection and movement of modules. The bioreactor system can be controlled and/or monitored by a controller that can individually identify and control each connected module and that can be adapted to collect signal data from sensors embedded in any of the modules.

Bioreactor, devices, systems and methods

Provided herein is a biohybrid elastomeric scaffold comprising a synthetic polymeric component and a biological polymeric component. The scaffold can be fabricated to have many different forms, non-limiting examples of which include a non-woven fibrous mesh or in a porous composite. Methods of use of the biohybrid elastomeric scaffolds in wound healing and tissue regeneration are also provided.

Biohybrid elastomeric scaffolds and methods of use thereof

A composition includes at least one biologically active agent covalently attached to a first polymerizing molecule that is adapted to undergo a free radical polymerization. The first polymerizing molecule retains the ability to undergo free radical polymerization after attachment of the bioactive agent thereto. The first polymerizing molecule is preferably biocompatible. The polymerizing molecule can, for example. be dihydroxyphenyl-L-alanine (DOPA) or tyrosine. The composition can also include a second component synthesized by reacting at least one core molecule having a plurality of reactive hydrogen groups with at least one multi-isocyanate functional molecule to create a conjugate including terminal isocyanate groups. The conjugate molecule is reacted with a second polymerizing molecule that is adapted to undergo a free radical polymerization. The second polymerizing molecule includes a reactive hydrogen to react with the isocyanate groups of the conjugate. The second polymerizing molecule retains the ability to undergo the free radical polymerization after reaction with the conjugate. In several embodiments, the first polymerizing molecule and the second polymerizing molecule are the same and dihydroxyphenyl-L-alanine (DOPA) or tyrosine.
