The present invention relates to a process for the production of interferon beta, and to an interferon beta composition having a unique glycosylation pattern.

Process for the preparation of glycosylated interferon beta

DNA encoding a soluble LDL receptor protein, is provided, as are vectors, cell lines and processes for the production of the protein, mutein or fragment. The soluble LDL receptor protein, and its muteins and fragments, are useful in protection of mammals against viral infections.

Soluble LDL receptor, its production and use

A soluble LDL receptor protein is provided. It can be isolated from cells that have been treated with an interferon, isolated from the urine of healthy human individuals or produced by recombinant techniques. The soluble LDL receptor protein is useful in protection of mammals against viral infections.

A cDNA encoding a new interferon-gamma (IFN-.gamma.)-binding protein is disclosed. Methods and products for the recombinant production of the protein are provided, and specific antibodies are also described.

Recombinant production of interferon-gamma binding proteins

A soluble LDL receptor protein is provided. It can be isolated from cells that have been treated with an interferon, isolated from the urine of healthy human individuals or produced by recombinant techniques. The soluble LDL receptor protein is useful in protection of meals against viral infections.

Method of antiviral use of soluble LDL receptor

A serum free medium capable of supporting production of mammalian cell products is provided. The medium comprises a basic medium and a cell viability protection agent, insulin and thrombin or a thrombin receptor activator.

Analysis of [.sup.125 I]. interferon gamma cross-linked to its receptor on various human cells by SDS-PAGE revealed that there are at least three different types of human interferon gamma receptors. In WISH, HeLa, FS11 and the other tissue cells an Mr 90,000-105,000 receptor was found. In monocytes and in the myeloid cell line KG-1 an Mr 140,000 receptor was found while in Daudi lymphoblastoid cells an Mr 95,000-115,000 receptor was found. The various receptors were isolated from these cells by extraction followed by affinity chromotography on an immobilized interferon gamma column. The resulting purified preparations retained their original affinity for interferon gamma and were used for immunizing mice and subsequent development of highly specific antibodies.

There is provided a process for purification of human immune interferon (IFN-.gamma.) to homogeneity without appreciable loss of biological activity by the use of chromatography on controlled-pore glass, ultrafiltration and high performance cation exchange chromatography, resulting in essentially pure human interferon subtypes 21K and 26K, and there are provided said purified interferons and pharmaceutical compositions containing same.
