The present invention provides a process for the determination of a specifically bindable substance by incubation of the sample solution with at least three receptors R.sub.1, R.sub.2 and R.sub.3, of which R.sub.1 and R.sub.2 are bindable with one another and R.sub.3 is specifically bindable with the substance to be determined and measurement of the agglutination which takes place in the case of the reaction, wherein, as receptor R.sub.1, there is used a conjugate of a partner of a specifically binding pair P and of a substance S which corresponds to the substance to be determined or is a derivative thereof and has at least one epitope of the substance to be determined, as receptor R.sub.2 there is used a receptor which has at least two binding positions for P and as receptor R.sub.3 there is used a receptor which has at least two binding positions, of which at least one binds specifically with an epitope of the substance to be determined or of S. The present invention also provides a reagent for carrying out the above process.

Process and reagent for the determination of a specifically bindable

For the determination of a specifically bindable substance by incubation of the sample solution with at least two receptors R.sub.1 and R.sub.2, whereby R.sub.1 and R.sub.2 are capable of binding to each other and R.sub.1 is capable of specific binding to the substance to be determined and measurement of the agglutination which occurs in the reaction, a conjugate of one partner of a specific binding pair P and a component K capable of specific binding to the substance to be determined is used as the receptor R.sub.1 and a receptor which has at least two binding sites for P is used as R.sub.2.

Process for the determination of a specifically bindable substance

The present invention provides compounds of the general formula: ##STR1## wherein R is a trifluoromethyl radical or a cyano group. The present invention also provides processes for the preparation of these compounds, as well as reagents containing them. The present invention is also concerned with the use of these compounds as .beta.-galactosidase substrates, especially in a CEDIA system.

Hapten-biotin conjugates in which the hapten is linked with biotin via a spacer, which has 26 to 40 atoms in its chain and contains at least 5 heteroatoms, are novel and are suitable, in particular for use in a competitive homogeneous immunoassay in which the agglutination which occurs in the reaction is evaluated by turbidimetric or nephelometric measurements.
