Anti-carbohydrate antibodies are detected by (a) contacting an array of oligomannose-serum albumin conjugates immobilized on a substrate with an antibody-containing serum sample under conditions wherein TM10 antibodies bind the oligomannose of the conjugates at at least micromolar affinity; and (b) detecting resultant binding of specific antibodies of the sample to the oligomannose of the conjugates, as indicative of the anti-carbohydrate antibodies.

Compounds and methods for controlling the surface properties are described. Compounds of the invention can form radicals upon exposure to irradiation, which can then react with nearby molecules to alter the surface properties of various substrates. The invention can provide surfaces that are resistant to dewetting, surfaces that have immobilized molecules such as carbohydrates and polymers immobilized, and surfaces that have metals deposited on the surface. The invention can be utilized in a wide range of application, such as sensors, microreactors, microarrays, electroless deposition of metals, and the like.

This invention provides novel nitrocellulose-based or Hydrogel-based microarrays and methods of making and using them (1) to detect the presence of one or more agents in a sample, (2) to determine the amount of one or more agents in a sample, (3) to determine whether a subject is afflicted with a disorder, and (4) to determine whether an agent known to specifically bind to a first compound also specifically binds to a second compound. This invention also provides kits which comprise the instant microarrays. This invention further provides antibodies capable of specifically binding to a glycomer present both on the surface of a mammalian macrophage or intestinal epithelial cell, and on a bacterial cell. Finally, this invention provides diagnostic methods using the instant antibodies.

Pneumococcus polysaccharide-related vaccines

Disclosed are methods for probing the immunogenic sugar moieties of prostate cancer cells. The methods detect a number of glyco-epitopes that are highly and differentially expressed among prostate cancers of various Gleason grades. The glyco-epitopes exist on the surfaces of prostate cells. The methods also comprise the detection of autoantibodies in prostate cancer subjects. The antibodies bound to a glyco-motif of N-glycans that is normally 'cryptic.' This target is highly expressed in prostate cancers. Lectins and antibodies that detect these glyco-epitopes that expressed in prostate cancer tissues includelectin (EEL);Lectin-I (PTL-I);Lectin-I-A4 (GSL-I-A4);Lectin-I-B4 (GSL-I-B4);I agglutinin (SNA-I;-L (PHA-L;agglutinin (GNA);agglutinin (NPA);agglutinin (Jacalin); and mAb TM10 (IgM).

Prostate cancer glycan markers and autoantibody signatures

This invention includes the conception of T-independent conjugate-vaccines and its application in the induction of antigen specific IgA response. We demonstrated that 1) &agr;(1,6)dextran can elicit a markedly enhanced IgA response in T-cell free mice (20-50 fold higher than in normal mice); 2)co-injection of the molecule with other antigens can enhance the IgA response to the co-antigen; and 3)a dextran-Gag conjugate can elicit the Gag-specific IgA. Thus, the invention identified &agr;(1,6)dextran as a carrier molecule for producing the T-independent conjugates and as an adjuvant for the enhancement of IgA production. The T-independent property of these conjugates makes it especially useful in vaccinations against HIV and other infectious and non-infectious diseases associated with T-cell deficiency.
