Provided herein are methods and related compositions and kits for dissolution of platelet clumps formed in collected blood samples. The present methods, compositions and kits are useful for improving blood sample quality and blood assay accuracy in a lab setting, especially in preventing misdiagnosis such as pseudothrombocytopenia or pseudoleukocytosis.

Methods and compositions for reversing platelet clumping

A method of manufacturing a laminate microfluidic device is described herein. Also described is the microfluidic device manufactured via the method of the disclosure, as well as use of the device to perform an assay. The laminate device includes a substrate layer, an adhesive layer having a cured adhesive with one or more channels or recesses formed therein, and a top layer.

Microfluidic device and method of manufacturing the same

Provided herein are systems, devices, and methods for performing optical multianalyte detection in a sample, such as a human blood sample. The system processes sample in a single-use support pack that holds the blood sample and contains pipette tips, buffers, reagent preparation wells and a monolayer. Sample is transferred from the sample pack to a single-use disc consumable that contains microwells pre-filled with dried chemistries, plasma separation features, and a hematocrit channel. The instrument comprises a cell imager, an absorbance module comprising a spectrophotometer, a fluorescent laser scanning module, and a camera. The system uses only a small amount of blood from a single heparinized sample, to simultaneously provide results for a full panel of routine blood tests spanning elecochemistry, clinical chemistry, hematology, immunoassays and combinations thereof.

Optical multianalyte detection

A device for separating blood plasma from whole blood includes a first reservoir and a second reservoir. The first reservoir is configured to receive a sample of whole blood including red blood cells and includes a collection region and a constricted region. The second reservoir is fluidically connected to the constricted region of the first reservoir, such that, responsive to centrifugal force applied to the device, the sample of whole blood disposed within the first reservoir separates into a first fraction and a second fraction. The first fraction is located in the collection region and includes blood plasma from which substantially all red blood cells have been removed. The second fraction is located in the second reservoir and includes blood plasma and red blood cells that have been removed from the first fraction by the centrifugal force. The constricted region inhibits the second fraction from entering the collection region.

The invention provides seminal computational approaches utilizing data from non-rare cells to detect rare cells, such as circulating tumor cells (CTCs). The invention is applicable at two distinct stages of CTC detection; the first being to make decisions about data collection parameters and the second being to make decisions during data reduction and analysis. Additionally, the invention utilizes both one and multi-dimensional parameterized data in a decision making process.

Method of using non-rare cells to detect rare cells

Methods are provided for detecting 5T4-positive circulating tumor cells in a mammalian subject. Methods of diagnosing 5T4-positive cancer in a mammalian subject are provided. The methods of detection or diagnosis indicate the presence of 5T4-positive metastatic cancer or early stage 5T4-positive cancer.
