The present disclosure provides methods of producing hepatocytes from induced pluripotent stem cells. Further provided herein are methods of using the hepatocytes for the treatment of a liver disease.

Methods for the production of hepatocytes

Provided herein are methods for the production of antigen-specific effector T cells and NK cells from pluripotent stem cells which express a chimeric antigen receptor (CAR). Further provided herein are methods for the adoptive cell therapy by administering the effector T cells and/or NK cells provided herein.

Antigen-specific immune effector cells

Provided herein are methods for the efficient in vitro differentiation of somatic cell-derived pluripotent stem cells to hematopoietic precursor cells, and the further differentiation of the hematopoietic precursor cells into immune cells of various myeloid or lymphoid lineages, particularly T cells, NK cells, and dendritic cells. Provided herein are methods for the efficient in vitro maintenance, expansion, culture, and/or differentiation of pluripotent cells with disruption of the MeCP2 gene into various erythroid, myeloid, lymphoid, or endoderm lineages, particularly mature erythrocytes. The pluripotent cells may be maintained and differentiated under defined conditions; thus, the use of mouse feeder cells or serum is not required in certain embodiments for the differentiation of the precursor cells.

Generating mature lineages from induced pluripotent stem cells with MECP2 disruption

The present disclosure generally regards methods and compositions for providing multi-lineage hematopoietic precursor cells from pluripotent stem cells (PSCs). The PSCs comprise an expression construct encoding an ETS/ERG gene, GATA2 and HOXA9. Also provided are methods for providing hematopoietic stem cells capable of long-term engraftment in mammals, such as humans. Methods and compositions relating to the production of induced pluripotent stem cells (iPS cells) are disclosed. Provided are methods for generating mast cells from pluripotent stem cells in vitro. Methods are disclosed for the differentiation of pluripotent cells, such as iPS cells and/or human embryonic stem cells, into mast cells. The resulting mast cells may be used for various purposes including screening cells for drug development and research. Growth factors which may be included in culture media according to the present invention include stem cell factor (SCF), FLT-3 ligand, thrombopoietin (TPO), interleukin-3 (IL-3), and/or interleukin-6 (IL-6).
