A method for measuring concentrations of blood cell components is provided. The method comprises: obtaining a blood sample from a subject, the blood sample comprising at least one of red blood cells (RBCs), white blood cells (WBCs), and platelets (PLTs); mixing the blood sample with a non-lysing aqueous solution to form a sample mixture comprising a predetermined tonicity; passing the sample mixture through a flow cell; emitting light towards the flow cell; measuring at least one of an amount of light absorbed by the RBCs to obtain an RBC absorption value, an amount of light scattered by WBCs to obtain a WBC scatter value, and an amount of light scattered by PLTs to obtain a PLT scatter value; and determining a concentration of at least one of the RBCs, WBCs, and PLTs present in the sample mixture.

System and method for distinguishing blood components

A method for measuring concentrations of blood cell components is provided. The method comprises: obtaining a blood sample from a subject, the blood sample comprising red blood cells (RBCs), white blood cells (WBCs), and platelets (PLTs); mixing the blood sample with a non-lysing aqueous solution to form a sample mixture comprising a predetermined tonicity; passing the sample mixture through a flow cell; emitting light towards the flow cell; measuring an amount of light absorbed by the RBCs; measuring an amount of light scattered by WBCs, and PLTs; determining a concentration of each of the RBCs, WBCs, and PLTs present in the sample mixture from the measured amount of light absorbed by the RBCs and scattered by the WBCs and PLTs.

The enumeration and analysis of residual white blood cells in a sample of leukocyte-reduced blood products is conducted by forming a suspension of the leukocyte-reduced blood products with a sufficient amount of a lysing reagent. The lysing reagent comprises a buffer with a low molar concentration, and a non-ionic surfactant. The suspension of leukocyte-reduced blood products and the lysing reagent is incubated for a sufficient time at a temperature sufficient to selectively lyse the platelets and red blood cells without damaging the white blood cells. The white blood cells of the lysed blood products are then contacted with a suitable dye to stain the white blood cells and the number of stained white blood cells is measured. The lysing reagent is free of harsh organic solvents which can damage the plastic components of automated clinical analyzers.

The present invention describes semi- and fully-automated methods and reagents therefor for the assay and analysis of body fluid samples, particularly non-blood samples. The methods and reagents are especially useful for the assay and analysis of cerebrospinal fluid (CSF) samples. The reagent compositions sphere and fix all cells in the sample in suspension. Reported results can include red blood cell (RBC) and white blood cell (WBC) counts, WBC differential values, cell-by-cell volumes and dry-mass concentrations.

The invention provides a new single channel, single dilution method and system for identifying, analyzing and quantifying the cellular components of whole blood using a single channel, rather than multiple channels, of an automated hematology analyzer utilizing flow cytometry and the detection of the light scattered and absorbed by each cell. The single channel utilized in the method was previously known and used only for red blood cell and reticulocyte analysis. The method involves the use of an organic dye in the reagent solution for staining the nucleic acid of reticulocytes, including reticulated platelets, and white blood cells in the sample. The single channel method developed and described is particularly useful for determining white blood cell counts and assessing parameters of a whole blood sample, for blood samples from both human and non-human mammals. The single channel method of the invention employs only one blood diluent reagent composition that is mixed with the blood sample aliquot for analysis and, optimally, a sheath/rinse reagent, thus making it simpler and easier to use. In addition, the reagent compositions of the method are more benign to the white blood cells than previous reagents, which contained various lytic surfactants for removing red blood cells, for example. The apparatus for performing the method provides an economical, streamlined and space-saving analyzer for carrying out complete blood cell analysis on a mammalian whole blood sample.

Single channel, single dilution detection method for the identification and quantification of blood cells and platelets in a whole blood sample using an automated hematology analyzer

The invention describes novel methods for calibrating or standardizing flow cytometry instruments using synthetic polymer particles or beads having physical properties which provide advantages for their use in such instruments. The polymer particles or beads employed in the calibration methods are spherical, have an average particle diameter of from about 1 to about 8 microns, a relatively narrow particle distribution, and a low refractive index, i.e., from about 1.35 to about 1.45. The particles are safe, stable for long time periods and are particularly suitable for employment as hematology analyzer calibration substances. The invention provides an advantageous improvement over the types of standards currently used to calibrate a variety of instruments that rely on light scatter combined with other parameters to identify and measure biological particles or cells, as well as nonbiological particles. In particular, using the calibration/standardization method and polymer particles according to the present invention allows hematology instruments to be calibrated and standardized for subsequent accurate and reliable determinations of red blood cells, reticulocytes, white blood cells and platelets in whole blood samples.

The invention provides a very fast and reliable method and reagent composition for determining reticulocyte and erythrocyte counts, identification and characterization, as well as platelet counts, in a whole blood sample using fully automated hematology analyzers. The reagent composition includes a zwitterionic surfactant as sphering agent for reticulocytes and erythrocytes, an organic cationic dye, e.g., Oxazine 750, for staining the reticulocytes, and buffer solution components for maintaining a reagent pH of about 7.2 to 7.8, and an osmolarity of about 292.+-.5. On the basis of the particular pH, ionic strength, and dye concentration, the reagent composition of the present method improves upon previous methods, thus allowing fully automated blood sample analyses to be completed in less than about 30 seconds, with only about a 20 second incubation of sample in the reagent solution before analysis. With such a rapid incubation time, the reagent mixture is passed through the sensing region of a flow cytometer, and the light scattered and absorbed by each cell is rapidly measured to provide accurate cell determinations and counts. The volume and hemoglobin concentration and content of each erythrocyte or reticulocyte are determined. Mean cell volume, mean corpuscular hemoglobin concentration, and mean cell hemoglobin of the reticulocytes and/or erythrocytes are calculated from the measured cell-by-cell volume and hemoglobin concentration.
