Liposomes containing tau peptides, preferably phosphorylated tau peptides, and conjugates containing tau peptides, preferably phosphorylated tau peptides, conjugated to an immunogenic carrier are described. Pharmaceutical compositions and uses of the liposomes and/or conjugates for treating or preventing a neurodegenerative disease or disorder, such as Alzheimer's Disease, are also described.

Compositions of phosphorylated tau peptides and uses thereof

Methods for inducing an immune response against tau protein in a subject suffering from a neurodegenerative disease, disorder or condition, such as Alzheimer's Disease, are described. The methods include administering a liposomal priming composition containing tau peptides, preferably phosphorylated tau peptides, and a conjugate boosting composition containing tau peptides, preferably phosphorylated tau peptides, conjugated to an immunogenic carrier.

Essentially pure compounds of the formulas (I) to (XX) are provided. Compositions and methods for enhancing or stimulating an immune response are also provided. The compounds, provided are advantageous in that the compounds are essentially pure and free from contaminants encountered when such compounds are purified from natural sources.

The present invention comprises novel compositions and methods for eliciting high immune responses, of great specificity yielding conformationally sensitive antibodies. These antibodies recognize specific epitopes on a wide variety of antigens including but not limited to, amyloid protein, prion protein, Pglycoprotein. The novel compositions of the invention comprise supramolecular antigenic constructs generally comprising a peptide sequence, covalently attached to pegylated lysine resulting in modified and enhanced peptide presentation. The unique modification methodology of the present invention is applicable to a variety of peptides and can ultimately be employed in therapeutic formulations and vaccines for diseases and disorders such as Alzheimer's disease. 

Antibodies obtainable using supramolecular constructs

The present invention relates to method for preparing liposome-based constructs comprising a peptide, particularly an antigenic peptide, of interest modified through hydrophobic moieties reconstituted in liposomes and to the antigenic constructs obtained with said method. The invention further relates to the use of said constructs for the therapeutic and diagnostic use in the treatment of diseases and disorders, which are caused by or associated with proteopathy such as Alzheimer's Disease.

Essentially pure compounds of the formulas (I) to (XXV) are provided. Compositions and methods for enhancing or stimulating an immune response are also provided. The method and constructs disclosed and claimed herein allow to control peptide conformation by modulation of the peptide lipidation pattern, spacer and liposome composition, or via co-administration with small molecules. The invention relates to an immunogenic composition which comprises, firstly, a carrier and, secondly, as antigenic structure, conjugates comprising all or part of the amino acid sequences of at least one peptide derived from an extracellular loop of the P-170 protein, each peptide being combined with several molecules of fatty acid containing a carbon chain of between C12 and C24 so as to allow, under suitable administration conditions, the induction of anti-P-170 antibodies.
