A method employing gel electrophoresis and optical imaging techniques to measure the amount of biomaterial that attaches to specified locations on a detector slide such as a bioarray or biochip.

Imaging electrophoresis system

In one embodiment, an apparatus is provided for testing for the presence of analytes in a sample. The apparatus comprises a source of light directed at the bottom surface of a substrate to achieve total internal reflection and to generate an evanescent field. An array of capture elements is immobilized on the top surface of the substrate. The bottom surface of the substrate is configured as a sawtooth (in cross section), the 'teeth' aligned with the rows or the columns in the array. The outer faces of the sawtooth 'prisms' are non-parallel to the substrate top surface, and specific requirements are imposed on the prism light-entrance face, and the substrate thickness and refractive index.

Ellipsometry systems for imaging binding events between analytes in a sample and an array of ligands in an evanescent field generated by a beam of light reflected from the plane of the ligands is adapted to a multiwell plate structure in common use. In one example, a film of prism arrays is affixed to the underside of the plate with each prism array located in registry with a well and ligand arrays being immobilized on the (planar) bottom surface of the wells. The prism array may be formed in a film and juxtaposed with the bottom surface of the plate or a prism array can be made integral with the plate bottom of a multiwell plate.

Apparatus and method for performing ligand binding assays on microarrays in multiwell plates

An apparatus and method for real time, label-free imaging and quantitation of binding events at an array of positions are provided. Total internal reflection from a planar side wall of a well of a multiwell plate is used to create an evanescent field in the plane of a pattern of ligands immobilized on the wall. Embodiments include imaging and multiple analyte detection and quantitation of a single wall of a single well as well as the simultaneous imaging and multiple analyte detection and quantitation of a number of wells.

Image data is acquired, processed, and/or displayed in accordance with an embodiment of the present disclosure to display, monitor, and/or demonstrate the progress of an experiment substantially in real-time and with high sensitivity. In one embodiment, at least one time-resolved value of spatially distributed polarization change data is provided and displayed. Advantageously, real-time processing and display of data is provided such that discussion and collaboration about the experiment may occur, time-resolved data is not lost, and resources are not wasted.

Image acquisition, processing, and display

An imaging method and apparatus are provided, including a light source emitting a polarized light beam, and an optical assembly including a control layer and/or a light reflection surface. The control layer advantageously allows for control over the properties of a generated evanescent wave to optimize an image of a specimen array within the evanescent wave. The light reflection surface includes coupling means used to couple a receptor/capture agent advantageously allowing for flexible control over receptor specific regions.

Imaging method and apparatus

Methods and apparatus for scanning of a microarray to provide an image of the microarray are disclosed. Data related to change in polarization state of a scanning light beam for individual points or lines or other portion of the microarray are collected and processed to image the microarray at high resolution and speed.

Microarray scanning

Apparatus for acquiring an image of a specimen comprising a cassette having an optical portion holding a specimen array on a TIR surface and being removably matable to a processing portion having a polarized light beam source and a processing polarization-sensitive portion to image the spatially distributed charges in polarization of the specimen array. In one form the array optical portion comprises a transparent slide having a bottom surface with first and second gratings located to direct polarized light to the TIR surface and to direct light reflected by that (TIR) surface to an imager, respectively. The apparatus may include a flow cell integral with the optical portion as well as means for selecting the direction and wavelength of the polarized light.
