Disclosed herein are methods and compositions for the identification of viability enhancing cell features and substrate features as they relate to post-cryopreservation survival of substrate seeded cells. Embodiments of the present invention further involve identification of cell features to manufacture a supernatant that is useful for cell culturing and treatment of various diseases.

Disclosed herein are methods and compositions for the cryopreservation of stem cells, such as stem-cell derived retinal pigment epithelial cells, that have been seeded onto and cultured on a substrate, such as a polymeric substrate. Such cryopreserved stem cells are useful for cell therapies, such as treatment of ocular damage or disease.

The production of high quality retinal pigmented epithelium (RPE) cells is necessary for research and potential therapeutic uses. Especially desirable are methods for the production of RPE cells using xeno-free culture conditions. Disclosed herein are novel methods for the production of RPE cells from pluripotent cells with high yields, including xeno-free production methods. Also provided are methods of efficiently isolating RPE cells from cultures containing heterogeneous cell types, allowing for substantially pure RPE cell cultures to be established. Additionally, novel methods for the cryopreservation of RPE cells are provided.

A method of treating ocular disorders (such as, for example, proliferative vitreoretinopathy or PVR) associated with replicating ocular cells by transfecting replicating ocular cells in vivo with a polynucleotide encoding an agent which is capable of providing for the inhibition, prevention, or destruction of the growth of the replicating ocular cells upon expression of the agent. The agent may be a viral thymidine kinase, and the polynucleotide encoding the agent may be contained in a retroviral vector. Once the replicating ocular cells are transduced with the retroviral vector, the patient is given a chemotherapeutic or interaction agent, such as ganciclovir, which kills the transfected replicating ocular cells.
