This invention relates to novel human polynucleotides and variants thereof, their encoded polypeptides and variants thereof, to genes corresponding to these polynucleotides and to proteins expressed by the genes. The invention also relates to diagnostic and therapeutic agents employing such novel human polynucleotides, their corresponding genes or gene products, e.g., these genes and proteins, including probes, antisense constructs, and antibodies.

A human gene encoding a novel mitogen-activated (MAP) kinase kinase, MKK7, provides methods and reagents for treating a variety of MKK7-mediated disorders, such as ischemic heart disease, kidney failure, oxidative liver damage, respiratory distress syndrome, heat and radiation burns, septic shock, autoimmune disorders, and inflammatory diseases. MKK7 reagents can also be used to induce or prevent apoptosis.

The present invention generally relates to novel GRB2 associating proteins and nucleic acids which encode these protein. In particular, these novel proteins possess inositol polyphosphate 5-phosphatase and phosphatidylinositol 5-phosphatase activities, important in growth factor mediated signal transduction. As such, the proteins, nucleic acids encoding the proteins, cells capable of expressing these nucleic acids and antibodies specific for these proteins will find a variety of uses in a variety of screening, therapeutic and other applications.

Novel polypeptides called signaling inositol polyphosphate 5-phosphatases are described called SIP-130, SIP-125, and SIP-N. SIP-130 is capable of binding to aPTB domain of SH2 and collagen containing protein (SHC). Also provided are polynucleotide sequences encoding the novel polypeptides, as well as vectors and host cells containing the polynucleotides. Further provided are modulators including agonists and antagonists of the novel polypeptides for use as therapeutics, including antibodies, polypeptides, small molecules, and polynucleotides and methods of using the therapeutics in treatment of diseases associated with abnormal cell growth. Methods of making the polypeptides, polynucleotides, vectors, host cells, antibodies, and small molecules are also provided. Gene delivery vehicles including SIP and SIP activity-modulating polynucleotides are described.

Signaling inositol polyphosphate 5-phosphatases (SIPs)

A novel kinase has been identified which phosphorylates I.kappa.B. Reagents which inhibit this kinase can be used as therapeutic tools to inhibit inflammation. The kinase can also be used as a target for drug screening to identify anti-inflammatory compounds.
