Novel DNA constructs are provided which may be used as molecular probes or inserted into a plant host to provide for modification of transcription of a DNA sequence of interest in cotton fiber, particularly in very early fiber development. The DNA constructs comprise a cotton fiber transcriptional initiation regulatory region associated with a gene which is expressed in cotton fiber.

A method of producing human serum albumin (HSA) in monocot plant seeds, by transforming a monocot plant cell with a chimeric gene containing a promoter from the gene of a maturation-specific monocot plant storage protein, a first DNA sequence operably linked to the promoter and encoding a signal sequence, and a second DNA sequence linked in translation frame with the first DNA sequence and encoding HSA, where the first DNA sequence and the second DNA sequence together encode a fusion protein containing an N-terminal signal sequence and the HSA; growing a monocot plant from the transformed monocot plant cell for a time sufficient to produce seeds containing the HSA; and harvesting the seeds from the plant.

A method is provided for transforming solanaceous plants to express DNA sequences interest from the plant cell plastid. The improved method allows the transformation of solanaceous plant tissue which is not obtained from tobacco with DNA constructs. Such DNA constructs comprise, in the 5&prime; to 3&prime; direction of transcription, a promoter region functional in a plant plastid and a DNA sequence of interest. The method can be utilized in the transformation of solanaceous plants, such as potato and petunia. The invention further provides constructs and methods for the expression of green fluorescent protein from the plant cell plastid.

The present invention is directed to the modification of reserve polysaccharides in plants. Specifically, it has been found that host plants can be successfully transformed with a nucleic acid sequence capable of expressing a chimeric reserve polysaccharide modification enzyme gene sequence which will synthesize novel reserve polysaccharides in plants or convert the transformed plant's endogenous starch reserves to novel starch degradation products.

Provided are constructs and methods for expressing herbicide tolerance genes in plastids of plant cells. Constructs include the components of a promoter functional in a plant plastid, a DNA sequence which is capable of conferring tolerance in a plant cell to at least one herbicide compound when said DNA sequence is transcribed in plastids of said plant cell and a transcription termination region. Herbicide tolerance is produced by transforming plastids with the constructs of the invention and growing plant cells comprising the transformed plastids under conditions wherein the DNA sequence is transcribed and plant plastids and cells containing the plastids are rendered tolerant to applications of at least one herbicide compound.
