This invention provides a medical device for delivering volumetric quantities of a first and a second biochemically reactive fluid in which separate MBUs atomize the fluids. The device of the present invention includes a fluid delivery device for laparoscopically delivering fluids having two feed conduits for delivering fluid to feed ports of the mechanical breakup units, the mechanical breakup units having two feed ports for receiving fluid from the feed conduits and directing such fluid into spin chambers and funneling portions having a sloped sidewall for directing fluid from the spin chambers through an exit port. The device is suitably dimensioned for creating a spray of two fluids with different viscosities such as fibrinogen and thrombin in such a manner that the thrombin spray cone encompasses the fibrinogen spray cone at a distance of about one inch from the exit port with a 0.5 cc/second flow rate to avoid depositing unreacted fibrinogen on a surface. Other instruments such as optical fibers may be incorporated within the device.

Mechanical breakup unit for biochemically reactive fluid delivery device

This invention provides a medical device for delivering volumetric quantities of a first and a second biochemically reactive fluid comprising a first container having an opening, the first container being adapted to contain the first biochemically reactive fluid; a second container having a second fluid opening adjacent the first fluid opening, the second container being adapted to contain the second biochemically reactive fluid; a spray unit for separately atomizing the first and second biochemically reactive fluids into an aerosol with at least one energy source of a liquid energy, a mechanical energy, a vibration energy, and an electric energy; a fluid pressurizer for pressurizing the first and the second biochemically reactive fluids for delivery under pressure through the spray unit onto a surface; and wherein the first and second biochemically reactive fluids first mix on the surface.

Fibrin delivery device and method for forming fibrin on a surface

An in vitro tissue testing system especially useful for testing blood/tissue interactions. The system includes a tissue holding frame which forms a chamber between two flat pieces of tissue with a spacer plate therebetween. The spacer plate includes a reservoir aperture bordered on both sides by the opposed tissue pieces, and a mechanism for depressing portions of the tissue pieces inwardly into the chamber to propel blood from one side of the chamber to the other. The flow created simulates actual blood flow in a host system. The reservoir aperture may be hourglass shaped with circular end portions and a middle blood passageway. The actuating mechanism may include pistons for alternating contact with the tissue within the confines of the opposite reservoir end portions, thus propelling blood from one end portion to the other. The pistons may be driven by a plurality of eccentric cams on shafts coupled to a common drive motor. The holding frame may include two outer plates for sandwiching the tissue pieces against the spacer plate, the outer pieces having openings corresponding to the reservoir end portions, and the pistons reciprocate through the openings into contact with the tissue.

In vitro tissue testing system

A method for determining the activation level of platelets and other types of blood cells that undergo activation by ELISA and other immunoassay techniques includes the step of reacting a sample containing the blood cells in a liquid phase with an excess quantity of an activation-specific primary antibody prior to allowing the cells in the sample to bind to a solid surface. This differs from typical ELISA procedures, wherein the cells are bound to the surface of a plastic test tube or microtiter plate prior to addition of the primary antibody. It has been discovered according to the invention that binding the cells to a surface prior to reaction with the primary antibody changes the activation level of the cells, making the assay less accurate.

Standard platelet samples and method for preparation thereof

A process for the separation and isolation of AHF, fibronectin and von Willebrand's ristocetin cofactor from blood plasma by adding a sulfated mucopolysaccharide to blood plasma to a concentration of from about 0.05 to about 0.4 mg/ml of plasma, cooling the plasma to 0.degree. C.-15.degree. C., centrifuging the plasma to isolate a precipitate which forms and then removing the AHF-rich supernatant liquid from the precipitate. The precipitate is washed in a slightly basic solution of dilute salts and then dissolved in a buffered solution of chaotrophic salts of the Hofmeister series having an ionic strength from 0.01-1. Fibronectin is separated by chromotographic means from the dissolved precipitate which contains fibronectin and fibrinogen. The dissolved precipitate may also be applied to a glass or plastic column containing a 4% agarose gel (E. G. Sepharose 4B or Biogel A-15m) in a 0.25 molar solution sodium phosphate titrated to pH 7.0 with any base, preferably TRIS. The von Willebrand's ristocetin cofactor elutes in the void volume of the column, whereas the fibronectin-rich fraction elutes later. Both fractions may be collected and concentrated. in a preferred embodiment, 0.20 mg of sodium heparin per milliliter of human plasma is employed with the heparinized plasma solution cooled for 3 hours at a temperature of 2.degree. C.-4.degree. C.

Process for separation and isolation of AHF, von Willebrand's ristocetin

A process for the separation and isolation of AHF and fibronectin from blood plasma by adding a sulfated mucopolysaccharide to blood plasma to a concentration of 0.15-0.25 mg/ml of plasma, cooling the plasma to 0.degree. C.-15.degree. C. centrifuging the plasma to isolate a precipitate which forms and then removing the AHF-rich supernatant liquid from the precipitate. The precipitate is washed in a slightly basic solution of dilute salts and then dissolved in a buffered solution of chaotrophic salts of the Hofmeister series having an ionic strength from 0.01-1.
