Embodiments of the disclosure concern systems, methods, and/or compositions for cultivation of mammalian viruses, including at least humanand sapoviruses within the Caliciviridae family of viruses. The ex vivo culture systems include intestinal enteroids in combination with bile or a functionally active fraction or component thereof. In specific embodiments, the culture system is utilized to test inactivation compounds for therapeutic or environmental efficacy and to test contaminated comestibles and/or environmental entities for determination of the presence of infectious virus. Furthermore, antiviral compositions may be tested using systems of the disclosure, including drugs, small molecule inhibitors, and biologics such as neutralizing monoclonal antibodies.

Cultivation of human noroviruses

Embodiments of the disclosure concern systems, methods, and/or compositions for cultivation of mammalian viruses, including at least human noroviruses and sapoviruses within the Caliciviridae family of viruses. An electroconductive touchscreen glove including a thumb pocket and an index finger pocket separated therefrom by a gap and leaves a user's other fingers exposed. An electroconductive contact layer, disposed on each of an entire front end and tip of the respective thumb pocket and the index finger pocket, is contactable with and operationally communicable with a capacitive touchscreen of an electronic device, such as a cellular phone or an iPad&#xae;, for example, to permit the user's direct control the actions of the touchscreen. First and second zippers are disposed on the thumb and index finger pockets, respectively. A connector body, such as a linked chain, has first and second ends attached to a slider on the first and second zippers, respectively, to secure the pockets on the user's thumb and index finger and to prevent the pockets from getting separated and misplaced or lost during non-use of the glove.

Controlling the formation of crystalline hydrates in various fluid systems, most notably, gas and oil transmission pipeline systems by contacting the systems with certain polymers or polymers associated with solid particles. The polymers useful are chelating polymers capable of interacting with charged gaseous molecules such as carbon dioxide, by removing the carbon dioxide, or more practically by scavenging for the carbon dioxide, to prevent the methane or ethane hydrate structures from forming since they require carbon dioxide to stabilize their structures.

Controlling the formation of crystalline hydrates in fluid systems

Double-stranded cDNA was synthesized from nucleic acid extracted from Norwalk virus purified from stool specimens of volunteers. One clone was isolated from a cDNA library constructed in a pUC-13 vector after amplification of the cDNA. The specificity of this cDNA (pUCNV-953) was shown by hybridization assays. The cDNA reacted with post (but not pre-) infection stool samples from Norwalk volunteers and with highly purified Norwalk virus, but not with other common enteric viruses such as hepatitis A virus and rotavirus. Finally, the probe detected virus in the same fractions of CsCl gradients in which viral antigen was detected using a specific Norwalk virus radioimmunoassay, and particles were detected by immune electron microscopy. Single-stranded RNA probes derived from the DNA clone after subcloning into an in vitro transcription vector were also used to show that the Norwalk virus contains a ssRNA genome of about 8 kb in size. The original clone was also used to detect additional cDNAs which represent at least 7 kb of nucleic acid of the Norwalk genome. The availability of a Norwalk-specific cDNA and the first partial genome sequence information allow rapid cloning of the entire genome and of establishment of sensitive diagnostic assays. Such assays can be based on detection of Norwalk virus nucleic acid or Norwalk viral antigen using polyclonal or monoclonal antibodies to proteins expressed from the cDNA or to synthetic peptides made based on the knowledge of the genome sequence. Assays using proteins deduced from the Norwlk virus genome and produced in expression systmes can measure antibody responses. Vaccines made by recombinant DNA technology are now feasible.

Methods and reagents to detect and characterize norwalk and related viruses

Recombinant proteins and peptides of Norwalk virus are claimed. Also claimed are vaccines against Norwalk virus and methods of immunizing against Norwalk virus using recombinant Norwalk virus antigens and derivatives thereof. Also claimed are vaccines for non-Norwalk virus agents including a portion of the Norwalk virus capsid as a carrier.

Methods and reagents to detect and characterize Norwalk and related viruses

A composition for preventing or retarding the formation of gas hydrates during the transport of a fluid comprising water and a hydrocarbon through a conduit. The composition is a homopolymer of vinyl caprolactam, or copolymers thereof, having a low molecular weight in the range of 500 to 2500, which is made in a polymerization solvent, preferably a glycol ether, most preferably 2-butoxyethanol and also including a carrier solvent, preferably monoethylene glycol.

A process of making a homopolymer of vinyl caprolactam (VCL), and a copolymer with dimethylaminopropyl methacrylamide (DMAPMA), optionally including with one or more polymerizable monomers (X), in the composition range, by weight, VCL 99 to 30%, DMAPMA 1-70% and monomer X 0-30%, respectively, by suspension polymerization in an aqueous/alcoholic solvent medium, without requiring a protective colloid, which comprises charging vinyl caprolactam in an aqueous/alcoholic solvent mixture, wherein the weight ratio of alcohol to total monomers is &frac16; to 3, and if present, forming a solution of DMAPMA and optional monomers in water, feeding the solution into the VCL charge at a predetermined rate, periodically adding initiator to the reaction mixture, and polymerizing at about 40&deg; to 130&deg; C., and, if desired, neutralizing the product, to form a polymer product at about 5% to 40% solids, homogeneously dispersed in the solvent.

Process for making water soluble polyvinylcaprolactam polymers

A composition for preventing or retarding the formation of gas hydrates during the transport of a fluid comprising water and a hydrocarbon through a conduit. The composition is a copolymer of vinyl caprolactam and vinyl pyridine, and terpolymers thereof, with vinyl pyrrolidone, preferably including 70-97% by weight vinyl caprolactam. Suitably such polymers have a molecular weight in the range of 500 to 2500, and are made in a polymerization solvent, preferably a glycol ether, most preferably, 2-butoxyethanol; and also including a carrier solvent, preferably monoethylene glycol.
