The invention relates to gene expression regulatory sequences, specifically introns that act as enhancers of gene expression, the promoter and terminator sequences endogenously associated with these introns. Presence of these intronic enhancer sequences in proximity to promoter sequences leads to enhancement of gene expression. Methods of finding such new intronic enhancer sequences and using them to generate transgenic plants are also described.

Regulatory sequences for modulating transgene expression in plants

The present disclosure discloses polynucleotide sequences that can be used to regulate gene expression in plants. Terminator sequences fromandthat are functional in plants are disclosed. Nucleic acid molecules, recombinant expression constructs, plants and seed comprising these terminator sequences are further disclosed.

The present disclosure concerns methods and compositions relating to UXS polypeptides and/or nucleic acids encoding UXS polypeptides. In certain claims, the methods and compositions are of use to improve digestibility and/or ease of grain processing. Such improvements relate to a modulation in arabinoxylan and/or hemicellulose content in transgenic plants. Such plants can, for example, comprise one or more nucleic acid sequences that inhibit expression of one or more UDP-Xylose Synthase (USX) genes.

UDP-xylose synthases (UXS) polynucleotides, polypeptides and uses thereof

The present disclosure concerns methods and compositions relating to UXS polypeptides and/or nucleic acids encoding UXS polypeptides. In certain claims, the methods and compositions are of use to improve digestibility and/or ease of grain processing. The present invention provides compositions and methods for manipulation of plant arabinoxylose and plant xylan synthases. Compositions include novel nucleotide sequences encoding xylan synthases and biologically active variants thereof. The invention also includes the discovery xylan synthase activity is a property of CslA polypeptides. Further provided are methods for xylose manipulation using the sequences disclosed herein. One method comprises stably incorporating into the genome of a plant cell, a CslA encoding nucleotide sequence operably linked to a heterologous promoter and regenerating a stably transformed plant. Methods for enhancing digestibility of plants and improving gum production in plants are also provided.

The method comprises transforming a plant cell to comprise a nucleotide sequence operably linked to one or more of the regulatory sequences of the present invention and regenerating a stably transformed plant from the transformed plant cell.
