A microplate assembly comprising a multi-well microplate, a plurality of vent caps and a porous vent film. The microplate includes a frame that houses a plurality of open wells in a rectangular array. Vent caps mount on the microplate to seal and vent the wells. When the vent caps are coupled to the wells, an interior volume is formed in each well. The wells function as a vessel for liquid samples that occupy predetermined spaces within the interior volumes. Each liquid sample remains within its predetermined space for all orientations of the microplate assembly. The vent cap comprises an array of well inserts. Each well insert comprises a sealing plug and a vent tube. A flexible perforated web interconnects the well inserts to each other. The vent tubes are fixed to the sealing plugs and terminate in a vent. A barrier formed from a plurality of nested flaps resiliently mounts on the vent tube to partially cover the vent and help inhibit the evaporation and loss of the liquid samples.

Method for sealing and venting a microplate assembly

A microplate assembly comprising a multi-well microplate, a plurality of vent caps and a porous vent film. The microplate includes a frame that houses a plurality of open wells in a rectangular array. Vent caps mount on the microplate to seal and vent the wells. When the vent caps are coupled to the wells, an interior volume is formed in each well. The wells function as a vessel for liquid samples that occupy predetermined spaces within the interior volumes. The liquid samples remain within its predetermined space for all orientations of the microplate assembly. The vent cap comprises an array of well inserts. Each well insert comprises a sealing plug and a vent tube. The well inserts are interconnected by a perforated web. The sealing plugs form a seal at the mouth of the open wells. The vent tubes, which are fixed to the sealing plugs, terminate in a vent that communicates with the interior volumes outside the predetermined spaces occupied by the liquid samples. The vents permit the pressure within the interior volume to be equalized with the ambient pressure via a passage that runs through the vent tube and the sealing plugs. Material may be added to or removed from each well without removing the vent cap. The porous vent film, which has an adhesive coating, adheres to the vent caps while covering the passages, thereby inhibiting evaporation of the liquid samples.

The present invention relates to a method for the processing of molecules by determining one or more selection parameters for a plurality of molecules; selecting a compatible grouping of molecules based on the selection parameters to form a set; forming a mixture of molecules of interest from the set; and resolving the mixture to fractionate the molecules of interest. Described is a new enzyme, 31-O-desmethyl-FK-506 O-methyltransferase (FKMT) and its N-terminal amino acid sequence. This enzyme can specifically and preferentially methylate the C-31 hydroxy group in 31-O-desmethyl FK-506/FR-900506 and other 31-O-desmethyl analogs of FK-506/FR-900506 and related compounds such as 31-O-desmethyl immunomycin type molecules, also designated FK-520 and FR-900520 by Fujisawa. The enzyme can be isolated from a FK-506 producing microorganism Streptomyces sp. (Merck Culture Collection No. MA 6858) ATCC No. 55098. Employing the enzyme in its active form, supplemented with Mg.sup.+2 ion, in the presence of the methyl donor, S-adenosyl methionine (SAM), derivatives of 31-O-desmethyl FK-506, 31-O-desmethyl FK-520 and related 31-O-desmethyl compounds may be prepared.

Phenylalkyl-2,3-dihydrobenzofurans and analogs were prepared by (1) nucleophilic substitution involving an appropriately substituted 2,3-dihydrobenzofuranol (or 2,3-dihydrobenzopyranol) and a cinnamylhalide followed by reduction; or (2) by Wittig reaction involving a halomethyl derivative of 2,3-dihydrobenzofuran (or 2,3-dihydrobenzopyran and an aryl or a heteroaryl aldehyde followed by reduction. These compounds were found to be potent anti-inflammatory agents.
