A method for testing a microfluidic device includes interfacing a microfluidic device to a fluidic parameter testing system. The microfluidic device has an internal rotary valve and internal fluidic channels. Each channel has a port with a predetermined port position that the rotary valve is to align to in order to select any one of a plurality of reagents which flow through the channels. The rotary valve is rotated via the testing system to a plurality of rotary valve position of the rotary valve. A fluidic parameter of the microfluidic device is measured at each rotary valve position. The fluidic parameter is mapped relative to the rotary valve positions. It is determined from the mapping if the rotary valve aligns with each of the predetermined port positions for a flow of the reagents through the channels.

Method and apparatus to identify fluidic path positions

Devices for sequencing biopolymers and methods of using the devices are disclosed. In one example, such a device has a nanopore, a plurality of wells and fluidic tunnels to allow a biopolymer to translocate in the device. In some embodiments, the device may include integrated electronics or micro-electromechanical systems, such as valves, bubble generators/annihilators or pressure pulse generators, to actively control fluidic/ionic/electric flows in the device.

Isolation of cells in a nanopore sensor array

A circuit with electrical interconnect for external electronic connection and sensor(s) on a die are combined with a laminated manifold to deliver a liquid reagent over an active surface of the sensor(s). The laminated manifold includes fluidic channel(s), an interface between the die and the fluidic channel(s) being sealed. Also disclosed is a method, the method including assembling a laminated manifold including fluidic channel(s), attaching sensor(s) on a die to a circuit, the circuit including an electrical interconnect, and attaching a planarization layer to the circuit, the planarization layer including a cut out for the die. The method further includes placing sealing adhesive at sides of the die, attaching the laminated manifold to the circuit, and sealing an interface between the die and fluidic channel(s).

A method for scanning a microarray, including (a) providing a microarray to an optical scanner, wherein the microarray includes a surface having features having different target molecules; (b) scanning the surface in the x or y direction to acquire optical signals from the features at sequential regions of the surface, wherein the focus setting between the optical scanner and the surface is dynamically controlled in real time by repeatedly (i) adding a predetermined focal offset, thereby introducing an error in the focus setting, and (ii) adjusting the focus setting to reduce the error in the focus setting, thereby acquiring the optical signals from different regions at different degrees of focus such that a subset of the regions that are scanned have an introduced focus error; and (c) analyzing the optical signals that are obtained at the different degrees of focus to identify the different target molecules.

Dynamic autofocus method and system for assay imager

Biosensor including a device base having a sensor array of light sensors and a guide array of light guides. The light guides have input regions that are configured to receive excitation light and light emissions generated by biological or chemical substances. The light guides extend into the device base toward corresponding light sensors and have a filter material. The device base includes device circuitry electrically coupled to the light sensors and configured to transmit data signals. A passivation layer extends over the device base and forms an array of reaction recesses above the light guides. The biosensor also includes peripheral crosstalk shields that at least partially surround corresponding light guides of the guide array to reduce optical crosstalk between adjacent light sensors.

Biosensors for biological or chemical analysis and methods of manufacturing the same

An instrument includes a reagent management system. The reagent management system includes a plurality of reagent wells, each reagent well operable to contain a reagent of a plurality of reagents positioned therein. The reagent management system is operable to select a flow of reagent from one of the plurality of reagents. A flexible connection includes a laminate stack and includes a first flexible channel in fluid communication with the reagent management system. The first flexible channel is operable to route the flow of reagent therethrough. A flow cell includes a flow channel in fluid communication with the first flexible channel. The flow channel is operable to route the flow of reagent over analytes positioned in the flow channel. The flexible connection enables the flow cell to be moved by the instrument relative to a fixed reference point in the instrument.

An electrowetting-based droplet actuator includes top and bottom substrates, a droplet-operation gap between the top and bottom substrates, the droplet-operation gap including a gradually-reduced gap height in a direction of droplet flow when in use, and spaced electrodes embedded in the bottom substrate spanning a region thereof corresponding to the gradually-reduced gap height. A method includes gradually reducing a gap height in section(s) of a droplet-operation gap between top and bottom substrates of an electrowetting-based droplet actuator, the gradually reducing being in a direction of droplet flow when in use from a large-gap inlet to a small-gap outlet (relative sizes), the large-gap inlet being larger in height, the bottom substrate including spaced electrodes embedded therein spanning a region of the bottom substrate corresponding to the gradually reduced gap height, and moving dispensed droplet(s) of liquid in the direction of droplet flow using the spaced electrodes and an applied voltage.

A system includes a droplet actuator having a droplet-operation gap between top and bottom substrates, a reservoir(s) external to and coupled to the droplet actuator, the reservoir(s) sized for a large-volume fluid, and pressure source(s) external to the droplet actuator and coupled to the at least one reservoir. Operation of the system includes filling the reservoir(s) with a large volume of fluid(s), dispensing droplet(s) of the fluid(s) to the droplet-operation gap using the pressure source(s) as part of performing a droplet operation(s). Movement of the droplet(s) may be effectuated by activating the droplet actuator.

Droplet dispensing

A method includes engaging a well of a cartridge with a flow sensor of an instrument. The cartridge includes: a rotary valve including a rotatable port and a center port; the well in fluid communication with a channel, the channel including a channel port that the rotatable port is to align to in order to receive fluid from the well; and a flowcell including an inlet gasket in fluid communication with the center port. A source of pressurized air is connected to the flow sensor in order to establish a mass flow of air through a flow path. The flow path extends through one of the flow sensor, the channel, the rotary valve, and the flowcell. The mass flow of air through the flow path is measured with the flow sensor. It is determined if there is one of an air leak and an air blockage within the flow path.

In-situ fluidic inspection

Some embodiments provided herein include methods and compositions for the detection of target ligands on an array. In some embodiments, a capture probe specifically binds to a target ligand from a sample, the location of a bead comprising the capture probe in an array is determined, and the bead is decoded to identify the capture probe and the sample. In some embodiments, a barcode is indicative of a capture probe attached to a bead; and an index is indicative of a subpopulation of beads. In some embodiments, the barcode and the index are determined by sequencing.

Methods and compositions for identifying ligands on arrays using indexes and barcodes

An apparatus for directing fluid into and out of a fluidic device includes two or more fluid prime channels connected to a fluid inlet of the fluidic device, a flow control valve for each fluid prime channel to control flow between the fluid prime channel and the fluid inlet, one or more outlet channels connected to a fluid outlet of the fluidic device, and a flow control valve for each outlet channel to control flow between the fluid outlet and the associated outlet channel. An apparatus for delivering fluids to a fluid inlet includes a plate that is rotatable about an axis of rotation and a plurality of fluid compartments disposed on the plate, each compartment having a fluid exit port disposed at a common radial distance from the axis of rotation and positioned to align with the fluid inlet as the plate rotates about the axis of rotation.
