An aqueous gel composition comprising a) water, at least one polysaccharide and at least one high molecular weight polyethylene oxide, wherein the water content is at least 90% by weight of the composition, and b) a local anaesthetic agent or an analgesic agent, for use as a local anaesthetic or analgesic. The aqueous gel shows transparency, lubricity, stringiness, elongation, extensiveness, and cohesiveness while being devoid of taste and smell and non-tacky or non-sticky.

Aqueous gel composition and its use

A syringe () for mixing and ejecting an active pharmaceutical ingredient is disclosed. The syringe () comprises a syringe body () defining a first lumen (), a first plunger () arranged movably in said first lumen (), said first plunger () being hollow, thereby defining a second lumen () being separated from the first lumen (), and a second plunger () arranged movably in said second lumen (). A liquid diluent is contained in one of the first lumen () and the second lumen (), and an active pharmaceutical ingredient is contained in the other of the first lumen () and the second lumen (). The second plunger () is movable in such a manner that the active pharmaceutical ingredient and the liquid diluent are brought together, e.g. in the first lumen () or in the second lumen (), thereby causing the active pharmaceutical ingredient to be diluted by the diluent, and the first plunger () and/or the second plunger () is/are movable in such a manner that diluted active pharmaceutical ingredient can be ejected from the syringe (). The syringe () provides a single device for mixing and ejecting the active pharmaceutical ingredient. This reduces the risk of contamination, and the syringe () can be easily operated by an untrained user.

Syringe for mixing and ejecting an active pharmaceutical ingredient

A oral pharmaceutical composition for controlled release of an opioid from a pharmaceutical composition is described that comprises (i) an erodible matrix composition comprising a mixture of (a) a polymer or a mixture of polymers (b) an opioid, and optionally, (c) one or more pharmaceutically acceptable excipients; and (ii) a single, substantially insoluble in and impermeable coating having two openings exposing at least one surface of the matrix, and comprising one or more polymers. The composition exhibits controlled release of the opioid by erosion of the matrix and exhibits a zero order release profile wherein about 75% w/w of the opioid is released from the composition within 4-10 hours when tested in a dissolution test as described herein.

Morphine controlled release system

A syringe for mixing and ejecting an active pharmaceutical ingredient is disclosed. The syringe is designed in such a manner that it is ensured that substantially all of the mixed drug is ejected, and in such a manner that an active pharmaceutical ingredient can be lyophilized directly into the syringe. The syringe comprises a syringe body, a first plunger arranged movably inside the syringe body and a second plunger arranged movably inside the first plunger. In one aspect the second plunger comprises a first plunger part and a second plunger part, and the first and second plunger parts are adapted to cooperate to collapse the cavity of the first plunger. In a second aspect the cavity of the first plunger has a first diameter at a distal end and a second diameter at a proximal end, the first diameter being smaller than the second diameter.

A pharmaceutical composition for controlled release of an active substance is provided. The active substance is released into an aqueous medium by erosion of at least one surface of the composition. The composition comprises i) a matrix comprising a) polymer or a mixture of polymers, b) an active substance and, optionally, c) one or more pharmaceutically acceptable excipients, and ii) a coating.

Opioid polymer release system

A pharmaceutical composition for controlled release of an active substance, the composition being a matrix composition of: (a) a substantially water soluble or crystalline polymer, (b) an active substance, and optionally, (c) one or more pharmaceutically acceptable excipients having a water solubility of at least 1 mg/ml at ambient temperature. Described are controlled release oral pharmaceutical composition comprising an erodible matrix composition comprising a polyethylene oxide, a therapeutically effective amount of an opioid or a pharmaceutically acceptable salt, complex, solvate or anhydrate thereof, or a mixture thereof and, optionally, one or more pharmaceutically acceptable excipients, and a coating on the matrix composition, wherein the coating has at least one opening exposing a surface of an end portion of the matrix composition, thereby allowing controlled release of the opioid by erosion of the matrix surface, wherein the composition exhibits zero order release of at least 80% of the opioid contained in the composition. The matrix may have a cylindrical shape having a long axis and a conus-shaped end portion having a largest diameter and a smallest diameter, such that the surface area exposed increases during erosion of the conus-shaped end portion.

Layered pharmaceutical composition suitable for oral use in the treatment of diseases where absorption takes place over a large part of the gastrointestinal tract. The composition comprising A) a solid inner layer comprising i) an active substance, and ii) one or more disintegrants/exploding agents, one of more effervescent agents or a mixture thereof. the solid inner layer being sandwiched between two outer layers B1) and B2), each outer layer comprising iii) a substantially water soluble and/or crystalline polymer or a mixture of substantially water soluble and/or crystalline polymers, the polymer being a polyglycol in the form of one of a) a homopolymer having a MW of at least about 100,000 daltons, and b) a copolymer having a MW of at least about 2,000 daltons, or a mixture thereof, and iv) an active substance, which is the same as in said solid inner layer A), and layer A being different from layer B, the layered composition being coated with a coating C) that has at least one opening exposing at least one surface of said outer layer, the coating being substantially insoluble in and impermeable to fluids and comprising a polymer, and the composition having a cylindrical form optionally with one or more tapered ends, wherein the ratio between the surface area of one end surface of the cylinder and the length of the cylinder is in a range of from 0.02 to 45 mm.

Morphine polymer release system

A controlled release pharmaceutical composition for oral use comprising carvedilol. The composition releases carvedilol after oral administration to a mammal, including a human, in such a manner that a prolonged residence of carvedilol is obtained in the circulatory system compared with the known compositions of carvedilol. Furthermore, a composition according to the present invention makes available to the body a suitable plasma concentration of one or both of the enantiomeric species, namely R(+) and/or S(&#x2212;) carvedilol for obtaining the desired therapeutic effect.

Controlled release carvedilol compositions

A novel dosage form. The dosage form is presented in particulate form and before oral ingestion the particulate material is subjected to an aqueous medium, whereby it is converted to a semi-solid form by swelling or gelling of one or more of the components, especially of a gellan gum, of the particulate matter. The invention also relates to a vehicle for oral administration of one or more active substances, the vehicle comprising a gellan gum arranged in a configuration allowing optimal water diffusion so that upon addition of a predetermined amount of an aqueous medium, without the necessity of applying shear forces or other mixing forces, within a time period of 5 minutes or less swells and/or gels and the texture of the swelled vehicle being similar to that of a soft pudding and having a viscosity of at least about 10,000 cps as measured by a Brookfield Viscometer with a #4 LV spindle at 6 rpm and at 20-25&#xb0; C. In one embodiment of the invention, the particulate matter can be molded into a desired shape or pressed onto a dispensing unit such as a spoon.
