The present disclosure provides for rapid identification of mechanism of action (MOA) for drugs and toxins, and does so in a rapid (30 days or less) fashion. The methods use a combination of high throughput bioinformatics and pathway analysis that examine a wide variety of biological parametics.

The present invention relates to cellular proteins that are involved in toxicity and infection or are otherwise associated with the life cycle of one or more pathogens.

Mammalian genes involved in toxicity and infection

is a leading cause of antibiotic-associated infection in hospitals worldwide. There is a need for a vaccine tothat can target toxic proteins, or that can elicit adequate immunity to prevent infection or reduce the severity of infection. Modifiedtoxin A and B (TcdA and TcdB) proteins are described herein, which comprise mutations that reduce toxin A and B toxicity compared to the native toxin. The proteins described are highly similar to the native toxin of, but toxicity is reduced.
