A capillary array includes a light detection portion, a sample supply portion, a buffer solution supply portion and a voltage application portion which are necessary functions for electrophoresis, thereby, when assembling the capillary array into an electrophoresis apparatus, the same can be immediately used. Accordingly, a capillary array is provided which can be easily incorporated into an electrophoresis apparatus.

The flow rate of a solvent is reduced as a separation column and the separation performance is improved even under high-pressure conditions. There is provided a separation column including a monolith rod into which a sample and a mobile phase flow, the separation column comprising: a coating material coated on the outer circumference of a monolith rod; a support member into which the monolith rod coated with the coating material is inserted, and a rod fixing material fitted into or filled a gap between the coating material and the support member; wherein the upper end face of the rod fixing material is sealed, the upper end face being the inflow-side end when the separation column is assembled.

A method of electrophoresis, an electrophoresis apparatus and a capillary array in which a fluorescently labelled sample is supplied into capillaries in a capillary array constituted by a plurality of capillaries and including a sample supply portion, an electrophoresis medium supply portion and a detection portion while controlling the temperature of the capillary array through gas circulation, the sample is caused to migrate and separated in the capillaries through electrophoresis, and at the region where the capillaries are contacted or come close each other a plurality of capillaries are contacted to a solid body so as to dissipate heat generated from the capillaries to the solid body.

Method of electrophoresis, electrophoresis apparatus and capillary array

A microparticle measuring method according to the present invention, by which the number of fluorescent microparticles is counted and the fluorescent microparticles are analyzed, includes the steps of introducing fluorescent microparticles into a narrow flow path almost one after another; irradiating the fluorescent microparticles in the narrow flow path with excitation light; detecting a signal pulse produced by detection of a single photon of fluorescence generated by the irradiation with the excitation light; and recognizing existence of the fluorescent microparticle, starting from the number of signal pulses measured per predetermined standard period, and further includes the step of obtaining the number of signal pulses per standard period with a time interval shorter than the standard period. It further includes the step of counting successively the number of signal pulses generated in the predetermined standard period to recognize existence of the fluorescent microparticle, when the count value exceeds a predetermined threshold, and the kind of the fluorescent microparticles is estimated from the count value. Particularly by using microparticles having a diameter smaller than 0.1 .mu.m as label material, reaction efficiency of the label material is increased, stability of the binding with the material to be measured is raised, and the material to be measured can be detected with a high precision and a high sensitivity.

Method for measuring microparticles, quantitative measuring method

A body fluid sample is diluted with a diluent which comprises an aqueous solution containing bis(2-hydroxyethyl)-iminotris(hydroxymethyl)methane and boric acid to obtain a diluted body fluid sample, and the ion concentration in the diluted body fluid sample is determined with ion-selective electrodes, whereby the variation width of pH in the diluted body fluid sample can be held down to a narrow range. As a result the determination can be made without exerting adverse effects on the ion-selective electrodes, and errors in determination can be markedly reduced as compared with cases where the conventional diluents are used.

Determination of a plurality of species of antibodies or antigens is attained by a method which comprises forming a plurality of different kinds of reaction membranes each having a different species of antibody or antigen on an electrophoretic carrier, superposing these reaction membranes, optionally superposing a filter on the laminate of reaction membranes, inserting the laminate in an electrolyte, adding a plurality of different species of antigens or antibodies corresponding to the plurality of species of antibodies or antigens supported in the aforementioned reaction membranes, electrophoretically moving the added antigens or antibodies through the electrolyte and enabling them to react with the antibodies or antigens supported on the reaction membranes, and measuring the concentrations of either the antigens or antibodies resulting from the reaction or the antibodies or antigens supported in an unreacted form on the reaction membranes.

The degree of agglutination is measured in a magnetic manner instead of optical manner. Microparticles are composed of a magnetic material and the agglutinated condition is measured using a magnetometer. The sizes and distribution of agglutinated materials formed by the antigen-antibody coupling reaction can be correctly measured without affected by light-scattering materials such as proteins or blood cells contained in the specimen, or by absorbant such as pigment, or by fluorescent material. Therefore, the concentration of antigen is measured maintaining high precision.

An immunoassay method for measuring a concentration of an antigen for a short period of time by immobilizing an antibody over the whole zone of an effective supporting matrix for electrophoresis and fixing an antigen in a sample to be measured by electrophoresis for the antigen-antibody reaction between said immobilized antibody and said antigen.
