The disclosure pertains to methods and compositions for the rapid and efficient transformation of plants. The disclosure further provides methods for producing a transgenic plant, comprising (a) transforming a cell of an explant with an expression construct comprising (i) a nucleotide sequence encoding a WUS/WOX homeobox polypeptide; (ii) a nucleotide sequence encoding a polypeptide comprising two AP2-DNA binding domains; or (iii) a combination of (i) and (ii); and (b) allowing expression of the polypeptide of (a) in each transformed cell to form a regenerable plant structure in the absence of exogenous cytokinin, wherein no callus is formed; and (c) germinating the regenerable plant structure to form the transgenic plant. Methods and compositions for rapid plant transformation

Compositions and methods for modulating expression of gene products are provided. Compositions comprise suppression cassettes that comprise a convergent promoter pair operably linked to a silencing element that, upon expression, is capable of decreasing the expression of one or more target polynucleotides of interest. Compositions of the invention also include transformed plants, plant cells, plant tissues, and plant seeds. Methods of transformation and regeneration of plants comprising the novel constructs are provided. The methods find use in regulating gene expression, particularly genes associated with agronomic traits of interest. Further provided are promoter sequences, cells, plants, and vectors comprising these promoter sequences and methods of their use.

Compositions and methods for modulating expression of gene products

Compositions and methods for the alteration of the disulfide status of plant proteins are provided. Novel nucleotide molecules, comprising nucleotide sequences for thioredoxin h and NADPH-thioredoxin reductase, and the proteins encoded thereby are provided. The nucleotide sequences can be used to transform plants alone or in combination with other sequences, such as sequences encoding NADPH generating enzymes, to manipulate the thioredoxin h system and alter the protein disulfide status in plants. Transformed plants, plant cells, plant tissues, seed and grain are provided.

Compositions and methods for altering the disulfide status of proteins

Antisense expression of a stearoyl-ACP desaturase gene in sunflower results in more than a four-fold increase in seed stearate, and can enhance palmitate content as well. Thus, sunflower oil containing over 40 percent saturated fatty acids can be produced, which is desirable in the context of various products, such as coating fat, margarine, soap and shortening.

The present invention provides methods for increasing the level of preselected amino acids in seeds of plants, thereby enhancing the nutritional value of the seeds, by genetic modification. The present invention is particularly useful in increasing the methionine, lysine, and/or cysteine content in seeds of plants. Also provided, are isolated endogenous DNA molecules which encode soybean albumins. The present invention also provides an antibody which is capable of specifically binding to soybean albumins. The present invention further provides methods for isolating and purifying 2S albumins.
