A cell culture article includes a substrate having a polymer coating that is conducive to colony passaging of cells cultured on the coating. Example polymer coatings are formed from polygalacturonic acid (PGA), alginate, or combinations thereof. Cells cultured on the polymer coating can be separated from the substrate as a colony or layer of cells by exposing the polymer coating to (i) a chelating agent, (ii) a proteinase-free enzyme, or (iii) a chelating agent and a proteinase-free enzyme.

Proteinase-free coatings for colony passaging

A method for coating a surface of a cell culture article includes dissolving a polymer having a covalently attached polypeptide in an aqueous solution to produce a polymer solution. The polymer is formed from monomers selected to form a polymer having a linear backbone, wherein the polymer is crosslink free. The weight percentage of the polypeptide relative to the polymer conjugated to the polypeptide is sufficiently high to render the polymer conjugated to the polypeptide water soluble. The aqueous solution is substantially free of organic solvents. The method further includes (i) disposing the polymer solution on the surface of the cell culture article to produce a coated article; and (ii) subjecting the coated article to sufficient heat or electromagnetic radiation to attach the polymer conjugated to a polypeptide to the surface of the cell culture article.

A composition for forming a polymeric cell culture surface includes (i) a pre-polymer comprising a polymer backbone, a cationic moiety conjugated to the backbone, and a cross-linker moiety conjugated to the backbone; and (ii) a peptide-polymer comprising a polymer backbone and cell adhesive peptide conjugated to the backbone. Cross-linked coatings for cell culture that have a suitable amount of cell adhesive peptide and cationic moiety may be formed from the pre-polymer and peptide-polymer.
