The present invention relates generally to structural studies of the insulin binding site of the insulin receptor (IR) and the insulin-like growth factorreceptor (IGF-1R). More particularly, the present invention relates to the crystal structure of the low affinity insulin binding site of the IR ectodomain comprising the C-terminal region of the IR &#x3b1;-chain, as well as the corresponding region of IGF-1R, and to methods of using the crystal and related structural information to screen for and design compounds that interact with or modulate the function of IR and/or IGF-1R.

The present invention relates to the crystal structure of the insulin receptor ectodomain, to the nature of its N-linked glycans and to methods of using the crystal and related structural information to screen for and design compounds that interact with or modulate the insulin receptor and/or the closely-related insulin-like growth factor receptors or variants thereof.

The present invention relates to truncated EGF receptor molecules that exhibit increased binding affinities for EGFR ligands such as EGF and TGF1. The present invention also relates to methods of screening for EGF receptor ligands and methods of treatment which involve the use of these molecules.

Identification of the human insulin receptor binding site sequence of certain spatial molecular structures conforming to such binding site and of certain insulinomimetic sequences including the binding site sequence are disclosed.
