Among the various aspects of the present disclosure is the provision of a method of detection, treatment, and monitoring of cardiovascular disease or a foot wound by detection of a novel biomarker, Fatty Acid Synthase (FAS). Briefly, therefore, the present disclosure is directed to methods that allow for improved, noninvasive, and reliable diagnosis of these conditions, particularly in subjects suffering from Type 2 Diabetes (T2D).

Methods of detection and treatment for cardiovascular disease and foot wounds

The present invention provides methods and compositions for modulating certain metabolic processes and for treating a variety of disorders associated with metabolic syndrome, including insulin related disorders, ischemia, oxidative stress, atherosclerosis, hypertension, obesity, abnormal lipid metabolism, and stroke by administering an effective dose of a chloroquine compound. The invention also provides methods and compositions relating to administering an effective dose of a chloroquine compound in combination with at least a second pharmaceutically active ingredient or compound including an antihyperglycemic diabetes treatment, an antihypertensive agent, an antithrombotic agent, and/or an inhibitor of cholesterol synthesis or absorption.

Use of chloroquine to treat metabolic syndrome

Compositions comprising the peroxisome proliferator-activated receptor alpha (PPAR&#x3b1;)-ligand 1-palmitoyl-2-oleoyl-sn-glycerol-3-phosphocholine (16:0/18:1-GPC) are disclosed. These compositions may be used for the prophylaxis and treatment of PPAR&#x3b1;-related liver disorders including, but not necessarily limited to, fatty liver disease, to lower lipid and triglyceride levels, and to increase high density lipoprotein levels in animals. Foods modified with 1-palmitoyl-2-oleoyl-sn-glycerol-3-phosphocholine (16:0/18:1GPC) may be used to improve the metabolism of animals.

Use of an endogenous ligand for peroxisome proliferator activated receptor alpha to treat liver disorders

Disclosed is a 178 kDa glucose-inducible human fatty acid synthase (FAS) mRNA binding protein which has been purified to homogeneity and its binding element characterized. This large phosphoprotein binds to a novel repetitive element in the 3&prime; untranslated region (UTR) of the FAS mRNA. In particular, the binding has been mapped to a 37 nucleotide stretch within the first 65 bases of the 3&prime; UTR of mRNA. The binding protein is useful for mediating FAS expression, for regulating lipoprotein secretion and cell growth and for screening of test compounds for activity as inhibitors of FAS.
