A protein scaffold includes a plurality of EutM subunits and a multi-enzyme cascade. The multi-enzyme cascade includes a first enzyme attached to the first EutM subunit and a second enzyme attached to the second EutM subunit. The scaffold may be formed by a method that generally includes incubating a plurality of EutM subunits under conditions allowing the EutM subunits to self-assemble into a protein scaffold, attaching a first enzyme of a multi-enzyme cascade to a first EutM subunit, and attaching a second enzyme of the multi-enzyme cascade to a second EutM subunit. The scaffold may be self-assembled in vivo or in vitro. Each enzyme may be, independently of any other enzyme, attached to its EutM subunit in vivo or in vitro. Each enzyme may be, independently of any other enzyme, attached to its EutM subunit before or after the scaffold is assembled.

Self-assembling protein scaffolds and methods

This disclosure describes a cell that includes an engineered subcellular compartment that generally includes a proteinaceous shell that includes at least one bacterial microcompartment (BMC) polypeptide. In some embodiments, the engineered subcellular compartment can further include one or more targeted enzymes. We further describe methods of making and using such cells.

Engineered subcellular compartments

The present invention provides isolated polynucleotides and isolated polypeptides. The polypeptides of the present invention have isoprenoid wax ester synthase activity or isoprenoid acyl CoA-synthetase activity. The present invention also includes methods of using the polynucleotides and polypeptides of the present invention. For instance, the methods include producing biodiesel and producing wax esters.

Microorganisms for the recombinant production of resveratrol and other flavonoids

The invention relates to biotechnologically expressible, enzymically active recombinant porcine liver esterases, to a biotechnological method for the preparation thereof and to the use thereof in organic synthesis. The monomeric subunits of recombinant porcine liver esterase are truncated at their C-terminal end, compared with naturally occurring porcine liver esterase subunits. Moreover, it has proved to be an additional advantage to truncate the N-terminal end as well.
