The present invention provides antibodies comprising an antigen recognition domain that specifically binds to a metal chelate: mutant antibodies comprising a reactive site not present in the wild-type of the antibody, wherein the reactive site is in a position proximate to or within the antigen recognition domain; and methods of using such antibodies to diagnose and treat disease.

Radionuclide-labeled chelating agent-ligand complexes that are useful in medical diagnosis or therapy are prepared by reacting a radionuclide, such as .sup.90 Y or .sup.111 In, with a polyfunctional chelating agent to form a radionuclide chelate that is electrically neutral; purifying the chelate by anion exchange chromatography; and reacting the purified chelate with a targeting molecule, such as a monoclonal antibody, to form the complex.

Method for preparing radionuclide-labeled chelating agent-ligand

A method and system for localizing a diagnostic or therapeutic agent to an internal target site. The system includes (1) an epitopic compound, (2) a binding protein which is effective to bind specifically with the compound and capable of localizing selectively at the target tissue, when administered parenterally, and (3) a clearing agent which can bind to and cross-link the binding protein, to form a protein aggregate which is readily cleared from the subject's bloodstream. In practicing the method of the invention, the binding protein is administered to the subject parenterally, and allowed to localize at the target site, typically within 1-4 days. This is followed by a chase with the clearing agent to remove circulating, but not target-localized binding protein. When the epitopic compound is administered, binding of the compound to the localized binding protein, and rapid clearance of unbound compound by the kidneys, results in selective localization of the compound at the target site.

Method and system for administering therapeutic and diagnostic agents

A bleomycin conjugate for use in targeting a compound to a body tumor. The conjugate includes an oxidized bleomycin/cobalt(III) complex and the compound to be targeted joined to the complex through a monodentate cobalt/sulfur coordinate bond. Also disclosed are methods for preparing the conjugate, and for targeting the compound to a body tumor, for purposes of tumor radioimaging or therapy.

Bleomycin conjugates and method

Monoclonal Antibodies which are specific for a complex of a chelating agent and a metallic ion are described. The antibody has an association constant (K.sub.a) for the complex which is at least about ten times greater than the K.sub.a for the chelating agent alone or its complex with another metal.

Monoclonal antibodies against metal chelates

A copper chelate conjugate which is stable in human serum. The conjugate includes the copper chelate of a cyclic tetraaza di-, tri-, or tetra-acetic acid, a linker attached at one linker end to a ring carbon of the chelate, and a biomolecule joined at the other end of the linker. The conjugate, or the linker-copper chelate compound used in forming the conjugate, are designed for use in diagnostic and therapeutic applications which involve Cu(II) localization via the systemic route.

Macrocyclic bifunctional chelating agents

Leukocytes are radioimmunoimaged by injecting patients with an immunoreactive nonleukocidal conjugate of an anti-leukocyte antibody and a gamma-emitting radioactive metal chelate, waiting for the conjugate to localize on the leukocytes, injecting the patient with an antibody to the conjugate to clear the blood of background nonlocalized conjugate, and visualizing the leukocytes by scintillation scanning.

Reagent and method for radioimaging leukocytes

A simple method for making EDTA, ED3A or DTPA analogs from amide derivatives of alpha amino acids is disclosed. These EDTA, ED3A or DTPA analogs are useful chelating agents, and preferably are useful as bifunctional chelating agents which may be attached to biological molecules and which form physiologically stable chelates with a variety of metal ions.

Chelating agents and method

Bleomycin's affinity for tumorous tissue and its ability to complex with cobalt has lead to its use in vivo as a complex with .sup.57 Cobalt. This complex has given excellent tumor images and has been useful for diagnostic purposes. Unfortunately, .sup.57 Co has the undesirable property of a 270 day half-life. The present invention provides a bleomycin analog which has a powerful meal-chelating substituent covalently bound to and chemically modifying a bleomycin. However, the inclusion of this chelating substituent in the bleomycin analog does not significantly modify the in vivo biological properties thereof with respect to unmodified bleomycin. The inventive bleomycin analog forms stable chelates with a variety of metal ions, and is particularly useful when labelled with radionuclides having short half-lives. So labelled, the bleomycin analog may be utilized for diagnostic purposes.
