The present invention relates to methods of conducting cholinesterase inhibition assays. In one instance, the assays can be configured to determine the presence of inactivated and activated cholinesterases. Also described herein are microfluidic devices and systems for conducting such assays.

Endpoint detection of amplified nucleic acids

The present invention relates to modular assemblies and systems for conducting cell analyses. In particular embodiments, an assembly has micron-scale fluidic structures, thereby minimizing sample use, and possesses a modular architecture that facilitates disassembly of the apparatus. Thus, one such modular assembly facilitates on-chip cell growth and on-chip assays; and then, as needed, the assembly can be disassembled to provide direct access to the cells that were incubated and treated within the chip. Methods of making and using such assemblies, as well as systems thereof, are further described herein.

Modular assemblies and systems for cell cultures and methods thereof

The present invention relates to methods for detecting targets by employing a temperature control system with a microfluidic device. The system allows for non-contact heating by employing an infrared emitter. In some instances, the system can be used in conjunction with a centrifugal microfluidic device. Optionally, a mask can be implemented to provide selective heating of desired assay areas of the device.

Protein and nucleic acid detection for microfluidic devices

The various technologies presented herein relate to identifying whether an individual has taken, and/or is under the influence of, a restricted drug. A density separation technique is utilized, wherein a sample (e.g., blood, saliva, urine, etc.) which may include an analyte is exposed to a first plurality of beads having an analyte attached thereto, a second plurality of beads having a metabolite-specific antibody attached thereto, and a plurality of fluorophore-labelled analyte-specific antibodies. After incubation, any analyte in the sample (e.g., delta-9-THC) is bound to the fluorophore-labelled analyte-specific antibodies, any free fluorophore-labelled analyte-specific antibodies are attached to the analyte of the first beads, and any metabolite in the sample is bound to the second antibody. By applying centrifugal separation, the first beads move to a region which undergoes irradiation. If no fluorescence occurs, the sample includes the analyte; if fluorescence occurs, the sample does not include the analyte.

Detection of metabolites for controlled substances

The present invention relates to methods of detecting one or more targets of interest in a sample. In one instance, the target can be correlated to an active infection (e.g., by a virus and/or a bacterium). Methods can include treating the sample with a dissociation agent, thereby releasing the target of interest for more accurate detection (e.g., by use of a sedimentation-based centrifugal microfluidic devices). Also described herein are microfluidic devices and systems for use with a dissociation agent.

Detection of active and latent infections with microfluidic devices and systems thereof

Embodiments of the present invention are directed toward devices, systems, and methods for purifying nucleic acids to conduct polymerase chain reaction (PCR) assays. In one example, a method includes generating complexes of silica beads and nucleic acids in a lysis buffer, transporting the complexes through an immiscible fluid to remove interfering compounds from the complexes, further transporting the complexes into a density medium containing components required for PCR where the nucleic acids disassociate from the silica beads, and thermocycling the contents of the density medium to achieve PCR. Signal may be detected from labeling agents in the components required for PCR.

Method and apparatus for purifying nucleic acids and performing polymerase chain reaction assays using an immiscible fluid

Embodiments of the present invention are directed toward devices, systems, and method for conducting nucleic acid purification and quantification using sedimentation. In one example, a method includes generating complexes which bind to a plurality of beads in a fluid sample, individual ones of the complexes comprising a nucleic acid molecule such as DNA or RNA and a labeling agent. The plurality of beads including the complexes may be transported through a density media, wherein the density media has a density lower than a density of the beads and higher than a density of the fluid sample, and wherein the transporting occurs, at least in part, by sedimentation. Signal may be detected from the labeling agents of the complexes.

A system and methods for detection of a nucleic acid including forming a plurality of nucleic acid detection complexes are described, each of the complexes including a nucleic acid analyte, a detection agent and a functionalized probe. The method further including binding the nucleic acid detection complexes to a plurality of functionalized particles in a fluid sample and separating the functionalized particles having the nucleic acid detection complexes bound thereto from the fluid sample using a density media. The nucleic acid analyte is detected by detecting the detection agent.

Quantitative detection of pathogens in centrifugal microfluidic disks

Examples are described including measurement systems for conducting competition assays. A first chamber of an assay device may be loaded with a sample containing a target antigen. The target antigen in the sample may be allowed to bind to antibody-coated beads in the first chamber. A control layer separating the first chamber from a second chamber may then be opened to allow a labeling agent loaded in a first portion of the second chamber to bind to any unoccupied sites on the antibodies. A centrifugal force may then be applied to transport the beads through a density media to a detection region for measurement by a detection unit.
