Methods and materials for forming a three-dimensional (3D) structure in a material are described. An example method includes directing a liquid casting material into a mold cavity of a mold structure, where the mold cavity corresponds to a three-dimensional (3D) structure. The method further includes causing the liquid casting material to solidify within the mold cavity to form a solid structure of the casting material, removing at least a portion of the mold structure from the solid structure of the casting material, and forming a structural material around the solid structure of the casting material. The solid casting material is liquified within the structural material. The liquified casting material is evacuated from the structural material to form the 3D structure in the structural material.

Fabrication materials and processes useful to form structures in soft materials

A valve includes a body including an inner bore extending between a first port and a second port, a seat, and one or more restrainers and a disk that is moveable between the seat and the one or more restrainers such that a first pressure that is less than 1 pascal and applied in a first direction causes the disk to move from a first position towards a second position to permit fluid communication between the first port and the second port. A method for fabricating an electrode includes forming a 3D shell having a hollow interior and defining one or more openings, and directing an electrically conductive liquid through at least one of the one or more openings of the 3D shell, such that the 3D shell is at least partially filed with the electrically conductive liquid. The electrically conductive liquid can be caused to solidify within the 3D shell to form a solid electrode, or can remain a liquid to form a liquid electrode. The 3D shell can be formed having the one or more openings using a 3D printing process such as a two-photon writing system. The surface tension of the electrically conductive liquid aids in retaining the electrically conductive liquid within the 3D shell. The electrode can contact a tissue sample through one of the openings in the 3D shell.

Techniques for generating microtissues, including a micro-fabricated platform including at least one micro-well including a plurality of micro-cantilevers coupled thereto and surrounded by a plurality of ridges, each micro-cantilever including a cap at a terminal end thereof. The platform can be immersed in a suspension of cells. The suspension of cells can be driven into at least one micro-well, and the ridges can be de-wetted to remove excess suspension and isolate the suspension of cells in each micro-well. The cells can be driven in the suspension of each micro-well toward a top surface of the suspension, which can be polymerized to form a matrix. The cells can be cultivated to spontaneously compact the matrix such that the micro-cantilevers anchor and constrain the contracting matrix to form a band of microtissue that spans across the micro-cantilevers.

Methods of fabricating a substantially interconnected model vasculature, as well as compositions formed from such methods are provided. In some embodiments, the methods may comprise forming a non-woven fiber network comprising a plurality of fibers and a void space; backfilling the void space of the fiber network; and removing the fibers to form a substantially interconnected vascular network.

A method of patterning materials, such as proteins, on a contoured surface by depositing them onto protrusions on a surface, and a cell containment device that may be constructed by this method, are provided. The method may involve selectively depositing a material on a substrate including a contoured surface including protrusions and recesses. By applying a first fluid to the contoured surface and allowing the first fluid to distribute across only a portion of the contoured surface, a material may be deposited on the protrusions and not the recesses, or on the recesses and not the protrusions. Such a method may be used to selectively pattern cells or other materials on substrates.

The invention provides multisegmented, multifunctional magnetic nanowires for the probing and manipulation of molecules at the cellular and subcellular level. The different segments of the nanowire may have differing properties, including a variety of magnetic, non-magnetic, and luminescent behavior. Differences in surface chemistry allow different segments of a single nanowire to be functionalized with different multiple functional groups and/or ligands, giving the wire chemical multifunctionality.
