The present invention relates to compounds with high chemical stability and methods for inhibiting the pathological activity of extracellular histones in a subject. In particular, the invention relates to compounds with high chemical stability, uses thereof and methods for inhibiting or ameliorating extracellular histone mediated ailments (such as, for example, sepsis, systemic immune response syndrome (SIRS) and ischemia reperfusion injury (IRI)). More particularly, the invention relates to methods and uses of a polyanionic sulfated cellobioside modified with a small uncharged glycosidically linked substituent at its reducing terminus, wherein the presence of the substituent results in a molecule with high chemical stability without affecting the ability of the molecule to be effective in the therapy of extracellular histone mediated ailments. For example, the present invention relates to methods and uses of β-O-methyl cellobioside sulfate (mCBS) or a pharmaceutically acceptable salt thereof (e.g., mCBS.Na), in the therapy of a range of extracellular histone mediated ailments in subjects.

Compounds for treating and preventing extracellular histone mediated pathologies

The present invention relates to compounds with high chemical stability and methods for inhibiting the pathological activity of NETs in a subject. In particular, the invention relates to compounds with high chemical stability, uses thereof and methods for inhibiting or ameliorating NET mediated ailments (such as, for example, sepsis, systemic immune response syndrome (SIRS) and ischemia reperfusion injury (IRI)). The present invention is directed generally to chimeric proteins that can facilitate targeting of nanoparticulate carriers to antigen presenting cells, and to nanoparticulate carriers comprising these chimeric proteins. The invention is also directed to methods of internalizing an antigen in an antigen presenting cell, and methods of eliciting an immune response to an antigen in a subject, using the nanoparticulate carriers comprising the chimeric proteins.

Chimeric proteins

The invention relates to a method of inhibiting the cytotoxic activity of extracellular histones in a subject, comprising administering an effective amount of a polyanion to the subject. In particular the invention relates to a method for the treatment of patients who are suffering from sepsis and employs polyanions to rapidly form complexes with and thus neutralize or inhibit the cytotoxic activity of extracellular histone proteins, for example, those found in the blood circulation of sepsis patients.

The present disclosure relates to a composition for targeting dendritic cells. In particular, the present disclosure relates to a composition comprising: a) one or more antigens; b) an anti-DC-SIGN immunoglobulin single variable domain; and c) a carrier which carries a) and b). The disclosure further relates to formulations, compositions and devices comprising such anti-DC-SIGN molecules and their use as a medicament and in the treatment of cancer, suitably melanoma.

The compounds can also include substituted or unsubstituted cyclitol carbamides with the linker bond at the carbamide nitrogen. In other embodiments, the invention provides pharmaceutical compositions which include the compounds, and use of the compounds in the prevention or treatment in a mammalian subject of a disorder resulting from angiogenesis, metastasis, inflammation and/or coagulation/thrombosis

A method for the purification of human platelet heparanase using chromatographic techniques is described. The method comprises detergent-aided solubilization of a heparanase-containing fraction followed by purification of the heparanase therefrom by concanavalin A, Zn -chelating Sepharose, a Blue A or Reactive Red matrix, octyl agarose and gel filtration chromatography. Heparanase so purified has a molecular mass (Mr) of about 50 kDa and degrades both heparin and heparan sulfates.
