The present invention relates to novel affinity ligand-matrix conjugates comprising a ligand with the general formula (a) ##STR1## which ligand is attached to a support matrix in position (A), optionally through a spacer arm interposed between the matrix and ligand. The invention furthermore relates to these novel affinity ligand-matrix conjugates and the preparation and use thereof in the purification of proteinaceous materials such as e.g. immunoglobulins, insulins, Factor VII, or human Growth Hormone or analogues, derivatives and fragments thereof and precursors.

Two enzymes, one an acetylmorphine carboxyesterase (AMCE), the other a morphine dehydrogenase (MDH), have been isolated from bacteria. The AMCE degrades heroin to morphine and the MDH oxidizes morphine to morphinone, with the aid of a cofactor. More particularly, the invention pertains to a process for preparing a 6-ketone from morphine or a methyl or ethyl derivative thereof. The process comprises oxidizing morphine or an ether derivative in the presence of a morphine dehydrogenase and the cofactor NADP.sup.+. These reactions are used in detection of heroin (using the two reactions coupled together) or morphine. The enzymes can be incorporated in sensors for this purpose.

A device useful in diagnostics in which a biochemical species is attached to the surface of a sensor, especially to the gate of a field effect transistor. In such a 'BIOCHEMFET' it has been a problem to attach two or more biochemical species to the surface of the sensor, especially to the gates of a multi-gated FET. It has now been found possible to bond a group having a photoactivatable function covalently to the sensor surface, photo-expose the resultant modified surface selectively, e.g. through a mask, and bond the biochemical species, e.g. a hapten, antigen, antibody, lectin or enzyme, to the photoactivated function. In this way the biochemical species becomes attached in selected areas only. In view of the success of this technique realized under conditions of miniaturization, the invention makes possible 'printed circuits for proteins'. In addition to the device are the process of manufacture thereto, use for diagnostic purposes and a diagnostic kit of the device and a partner capable of binding to the biochemical species.
