A drug delivery device such as an oral dosage form (ODF) with a toxic or potent core encapsulated by a non-toxic region. The non-toxic region may be a region including multiple layers, coatings, shells, and combinations thereof, which provides protection to and isolation from the toxic or potent core. The drug in the toxic or potent core is incorporated into the dosage form via, for example, three-dimensional printing, as a solution, solubilization or suspension of solid particles in liquid, rather than by the more conventional handling and compressing of dry powder. This minimizes the likelihood of creating airborne particles of the toxic drug during manufacturing, hence controlling and minimizing the exposure of manufacturing personnel to the hazardous substance. Wet dispensing of the toxic or potent drug further provides greater bioavailability of the drug to the patient.

Method and form of a drug delivery device, such as encapsulating a toxic core within a non-toxic region in an oral dosage form

The invention includes a three-dimensional printing (3DP) machine that has at least one ultraviolet (UV) light source directed at powder or slurry being processed by the 3DP machine and/or directed at any of various surfaces or machine components, in a manner suitable to kill or damage microorganisms to a desired extent. The UV light sources may be either stationary or movable, and may be either pulsed or substantially continuous, and may operate in any desired sequence and timing with respect to other operations performed by the machine. The invention may include ultraviolet illumination in such a way as to interact with powder to sterilize or disinfect powder either while the powder is stationary or, alternatively, as powder is being moved from one place to another within the 3DP machine, or both. The invention also includes methods of performing three-dimensional printing using such illumination. The invention also includes the combination of other methods of sterilization or disinfection with the use of ultraviolet light. The invention also includes the use of ultraviolet light sources for a combination of polymerization/cross-linking in addition to sterilization/disinfection.

An engineered regenerative biostructure (erb) for implantation into a human body as a bone substitute, which includes an internal microstructure, mesostructure and/or macrostructure to provide improved bone in-growth, and methods for making the erb. Under one aspect of the invention, the biostructure has resorbable and nonresorbable regions. Under another aspect of the invention, the biostructure is constructed of hydroxyapatite, tricalcium phosphate and/or demineralized bone. Under yet another aspect of the invention, the porous biostructure is partially or fully infused with a resorbable, nonresorbable or dissolvable material.

Apparatus, systems and methods for use in three-dimensional printing are shown and described. Apparatus, systems and methods for use in three-dimensional printing

The rapid design and manufacture of biomedical devices such as implants, oral dosage pills and implantable pharmaceuticals employs electronic data and modeling transmissions via a computer network. Patient information and patient-specific radiological data is captured and transmitted via a computer network to a design and/or manufacturing site. A multi-dimensional digital model is created based on the radiological data and patient information. Communications interchanges between a clinical/diagnostic site and a design/manufacturing site permit modification of the digital model until approved. The approved digital model is converted into machine instructions to construct the biomedical device. Alternatively, the digital model is employed in a best fit selection of a biomedical device from a pre-existing set of biomedical devices or machine-instructions. A method and apparatus for dispensing and distributing a powder is disclosed that uses a rotary dispenser to dispense the powder by gravity onto a movable platen, the platen being movable to a position adjacent to a build bed where the powder can be distributed across the build bed using a spreader. The method and apparatus are also suited to dispensing of a plurality of powders used in build beds for solid free-form fabrication and three-dimensional printing. It is preferred that a powder contacting surface of the apparatus be coated with a coating selected from the group consisting of: an anodized finish, a hardcoat anodized finish, siloxane, baked enamel, electroplated enamel, electroless nickel, coated elastomer, laminated elastomer, dipped latex, sprayed latex, polyvinyl chloride, nylon, polytetrafluoroethylene, polyurethane, an epoxy resin, or a polyester.

A centrifugal pump for shear sensitive fluids includes a pump housing defining an interior volume. An impeller comprising at least two vanes is rotatably mounted in the housing. The impeller vanes, which preferably are airfoil-shaped, are spaced from the interior housing of the pump whereby to leave a gap between the vanes and the pump housing so as to permit a portion of the fluid in the spacing to flow smoothly over the top contour of the impeller vanes.

Pump for shear sensitive fluids

A power-assisted obturator system for penetrating a body cavity wall is provided. The system comprises an obturator having main shank with a tip on its distal end. The tip has relatively blunt dull cutting edges which are incapable of readily cutting through bodily tissue except when the tip is vibrated at ultrasonic or near-ultrasonic frequencies. The proximal end of the main shank is connectable to an ultrasonic power source, which provides power to vibrate the obturator tip at ultrasonic or near-ultrasonic frequencies thereby transforming the relatively blunt edges into cutting edges which simultaneously cut and coagulate tissue. In a preferred embodiment, the obturator system includes vibration feedback and control which terminates delivery of energy to the tip when penetration is sensed.
