Systems and methods for detecting and sequestering target analytes in a fluid sample. The system comprises a bulk specimen reservoir for receiving a liquid sample, along with paramagnetic materials operative to selectively bind to the target analyte of interest. A vertex is interconnected with the bulk specimen reservoir to define a specimen chamber assembly to which a magnetic chamber is deployed about the vertex portion thereof and operative to sequester the magnetic particles, as well as any analyte of interest bound thereto, within the interior of the vertex. Once sufficient time is allowed for mixing and allowing the magnetic capsule to sufficiently retain the magnetic particles within the vertex, the vertex is disconnected from the specimen reservoir and the paramagnetic materials retained therein analyzed to determine the presence of the analyte of interest.

Disclosed are methods to induce dislodgement of target prostatic cells from the prostate organ, collecting said cells, and subsequently examining the cell population. Such methods comprise the administration of an agent that facilitates the dislodgement of the target cells from within the prostate, which then migrate into the urethra. Exemplary agents include 5 alpha-reductase inhibitors. The cells induced to pass into the urethra are then collected non-invasively, such as through urine or semen samples. Such collection is further strategically calculated relative the administration of the agent so as to maximize the sample collection of the target cells of interest. The exfoliated prostatic epithelial cells are subsequently utilized for purposes such as detecting prostate cancer, predicting/measuring prostate tumor susceptibility to drug regimes, active surveillance of patients whose prostate biopsy results are negative, but continue to exhibit symptoms consistent with prostate cancer, and identifying false positive results associated with biomarker assays.

According to a preferred embodiment, a chromatographic medium is provided that is designed to be contacted with a test solution having activated magnetic particles such that the solution flows bilaterally thereacross. A magnetic field, generated by a magnet or electromagnet, is selectively applied to the medium which causes the charged particles to become substantially bound at a site on the medium specified by the position of the magnet, to thus form a captured line or zone. In one preferred embodiment, the magnetic field is applied at the site on the medium at which the test solution is contacted. The assay is multi-configurable and can be modified to suit a particular immuno-separation procedure or application.

The device includes a substantially planar, flexible article, a channel formed within the article, at least one fluid-receiving compartment formed within the article and fluidly connectable to the channel, and a solid phase movably positioned within the channel. The article may be a laminate, and in particular, a laminate formed from two substantially planar, flexible sheets laminated together.

Methods and apparatus for automated sample analysis are provided in which a plurality actuators are involved in moving a samples from one compartment to another, and appropriate reactants are combined with the sample in one or more of the compartments. The actuators are preferably contained in a device that also has a detector, data reduction capabilities, and a printer. Contemplated signal detectors include a photomultiplier tube, a photodiode, and a charge-coupled device. Steps contemplated to be performed automatically include aliquoting the sample, diluting the sample, contacting at least a portion of the sample with a reagent having a substantially selective binding towards the analyte. Contemplated reactants include sense and antisense nucleic acids, antibodies and antigens, solid-phases such as paramagnetic beads, reagents, other substrates, and wash solutions.

According to a preferred embodiment, the magnetic assay methods and systems incorporate a chromatographic medium, such as an assay test strip, that is designed to be contacted with a test solution having activated magnetic particles. A magnetic field, generated by a magnet or electromagnet, is additionally provided that if selectively applied to a chromatographic medium which causes the charged particles to become substantially bound at a site all in the chromatographic medium specified by the position of the magnets, to thus form a captured line or zone. To the degree of magnetic force applied to the medium may be selectively adjusted to vary the width or surface area of the capture line or zone. Additionally, in a preferred embodiment, capture lines may be formed while test strips are in motion along a stationary magnetized rail. Spectrophotometric analysis may be made at the captured lines and zones for qualitative and quantitative analysis of unidentifiable analyte. A multimode photometer consisting of an optical canopy and base that defines an optical chamber may be utilized for conducting spectral or multi-spectral analysis.

An improved enzymatic electrode and electrode module specifically adapted for use in a medical analyzer device is disclosed which permits rapid analysis of substances of interest contained within undiluted body fluids such as whole blood, serum and/or plasma. The enzymatic electrode is in fluid communication with an axially reciprocating probe which selectively transports either a quantity of a buffer aqueous solution or a calibrant aqueous solution presented at a wash cell or a body fluid specimen disposed within a sample cup to an active enzyme bearing membrane positioned adjacent the enzymatic electrode. The membrane comprises a composite membrane structure having a protective membrane layer adapted to prevent the passage of blood cells and other particulate or cellular substances therethrough as well as adjust the diffusion rate of the substance of interest desired to be measured into the membrane, an active enzyme layer adapted to convert the desired substance of interest to be measured into a detectable substance and a limiting membrane formed to prevent the passage of interfering low molecular weight substance therethrough. The membrane/reagent chemistry kinetics of the enzymatic electrode are specifically defined to enable a novel pseudo-rate/created peak measurement technique to be utilized which reduces measurement inaccuracies as well as signal data processing requirements. The membrane/electrode support structure and membrane/electrode solution flow path are specifically formed to permit rapid replacement of the composite membrane and aqueous solutions by non-professional personnel.

Enzymatic electrode and electrode module and method of use

A non-invasive method for measuring blood glucose concentration using glucose present at the mucosal surface of a living being is provided. The method includes the steps of positioning a glucose oxidase membrane on the mucosal surface, providing oxygen for peroxidative oxidation of the glucose present thereat, electrically measuring the amount of hydrogen peroxide product and calibrating that amount to determine blood glucose concentration. Also provided are electrode devices suitable for carrying out the method.
