The present invention provides antibodies directed against ICOS or a derivative thereof which neutralize ICOS engagement on Treg by inhibiting the fixation between ICOS and ICOS-L and abrogate proliferation of Treg induced by plasmacytoid dendritic cells. The present invention further provides antibodies directed against ICOS or a derivative thereof which induce IL-10 and IFN&#x3b3; production, induce CD4&#x2212; T cells proliferation, reduce Tconv proliferation, and increase the immunosuppressive function of Treg.

Antibodies directed against ICOS and uses thereof

Provided are antibodies that bind and inhibit CD73, are capable of increasing the proliferation of T cells in the presence of CD39-expressing B cells and ATP. The invention also relates to cells producing such compounds; methods of making such compounds, and antibodies, fragments, variants, and derivatives thereof; pharmaceutical compositions comprising the same; methods of using the compounds to diagnose, treat or prevent cancer.

The present invention provides novel materials and screening methods for identifying agonists and antagonists of cell receptors. Methods are disclosed for identifying agonists and antagonists using chimeric receptors comprising the extracellular ligand-binding domain of a first receptor fused with the transmembrane and intracellular domains of a second receptor containing an intracellular immunoreceptor tyrosine-based activation motif (ITAM).

Use of chimeric receptors in a screening assay for identifying agonists and antagonists of cell receptors

Dendritic cells play a critical role in antigen-specific immune responses. Materials and methods are provided for treating disease states, including cancer and autoimmune disease, by facilitating or inhibiting the migration or activation of antigen-presenting dendritic cells. In particular, chemokines are used to initiate, amplify or modulate an immune response. In one embodiment, chemokines are used to attract dentritic cells to the site of antigen delivery. An increase number of dendritic at the site of antigen delivery means more antigen uptake and a modified immune response.

Chemokines as adjuvants of immune response

Mammalian proteinases; related reagents and methods

A method is provided for generating human dendritic cells in vitro by treating CD34.sup.+ cells with tumor necrosis factor-.alpha. and interleukin-3 or with GM-CSF. The invention also includes cellular compositions of dendritic cells produced by this method. Dendritic cells of the invention can be used widely as components in many diagnostic and therapeutic systems, including improved mixed lymphocyte reactions for assaying tissue rejection, adoptive immunotherapy of cancer, adoptive immunotherapy of HIV and other viral infections, and SCID-hu mice for human antibody production.
