The present invention generally relates to novel bispecific antigen binding molecules for T cell activation and re-direction to specific target cells comprising a common light chain. In addition, the present invention relates to polynucleotides encoding such bispecific antigen binding molecules, and vectors and host cells comprising such polynucleotides. The invention further relates to methods for producing the bispecific antigen binding molecules of the invention, and to methods of using these bispecific antigen binding molecules in the treatment of disease.

T cell activating bispecific antigen binding molecules

The present invention relates to bispecific antibodies that specifically bind a T-cell activating antigen and a Tumor Antigen (TA), comprising a first Fab fragment and a second Fab fragment, wherein either the variable regions or the constant regions of the second Fab heavy and light chain are exchanged; and wherein the bispecific antibody does not comprise a Fc domain; methods for their production, pharmaceutical compositions containing said antibodies, and uses thereof.

Multispecific antibodies based on antibodies that share a common light chain are described herein. The multispecific antibodies include: modified heavy chains having, by domain exchange, a common light chain variable domain VL; and two modified light chains having, by domain exchange, variable heavy chain domains of a first antibody (VH) and a second antibody (VH), wherein one light chain is of kappa isotype and one light chain is of lambda isotype. The present invention also relates to methods for the manufacture of such antibodies and their use.

Multispecific domain exchanged common variable light chain antibodies
