This invention relates generally to mechanisms of gene expression in plants and more specifically to regulation of expression of genes in plants in a 'tissue-preferred' manner. Regulation of expression is achieved using a transcriptional regulatory unit capable of driving expression of genes within certain tissues of a plant. Said transcriptional regulatory unit are to be utilized for driving expression of genes effective in the control of insect or other pests which threaten plants.

Maize promoter sequence for leaf- and stalk-preferred gene expression

An expression control sequence which is intermediate in tissue specificity between constitutive and tissue specific is disclosed. This promoter is effective in expressing genes in the majority of tissues of corn, and can be used for effective expression of desired protein genes in plants.

Expression control sequence for general and effective expression of

A method for commercial production of aprotinin entails heterologous expression of the protein in plants, preferably at a level such that aprotinin accounts for at least 0.1% of the total extracted protein. An aprotinin-expressing plant also has potential of increased insecticidal resistance by virtue of producing the protein. A genetic map of the integration locus allows identification of plants derived from the transgenic plant. This approach also reveals genetic loci on a plant chromosome that support high levels of gene expression and can be used as site of integration for expression of other genes of interest.

Commercial production of aprotinin in plants

A method for commercial production of avidin entails heterologous expression of the protein in plants, in native conformation, at an expression level such that avidin represents at least 0.1% of total extracted protein. A genetic map of the integration locus allows for the identification of the avidin-expressing plant. Genetic loci on a plant chromosome are revealed that support high levels of avidin expression and that can be used as a site of integration for high level expression of other genes of interest.

Commercial production of avidin in plants

A novel DNA regulatory element that confers microspore-specific gene expression has been discovered, isolated, and characterized. The microspore-specific regulatory element can be used to control the expression of a foreign gene that disrupts the function of microspores. Thus, the control of pollen production can be achieved by using the microspore-specific regulatory element to produce male-sterile plants. Various methods can be used to restore male fertility in the F1 generation of such male-sterile plants. In addition, the microspore-specific regulatory element can be used to confer resistance to viral and insect pests.

Use of a microspore-specific regulatory element for production of virus

The Brassica ALS3 promoter is operably linked to a foreign structural gene to provide high level, and generally constitutive or tissue general expression of the structural gene in transformed plants. The Brassica ALS3 promoter exhibits a non-tissue-preferred mode of expression at a level comparable to, and in some cases higher than, the widely used CaMV 35S promoter. Accordingly, DNA constructs comprising the Brassica ALS3 promoter operably linked to any number of different gene coding regions can be used for constitutive and tissue-general expression of the gene in transformed plants. The Brassica ALS3 promoter is used to direct expression of agronomically important genes and selectable marker genes.
