A family of related intracellular skin type antifreeze polypeptides and corresponding coding nucleic acids are provided. These are the first skin type intracellular antifreeze polypeptides and coding nucleic acids ever reported. The polypeptides are naturally expressed in the skin of Winter Flounder, and skin specific promoters are also provided. The polypeptides are used to make cells cold-resistant, and to improve the palatability of cold foods and liquids. Cold resistant eukaryotes and prokaryotes, including plants, animals and bacteria are made using the skin-type intracellular antifreeze polypeptides and nucleic acids.

An 'all fish' chimeric gene construct suitable for gene transfer for commercially important fish comprises the antifreeze gene (AFP) promoter fused to the desired gene sequence which is incorporated into fish embryos. The desired gene sequence is expressed in the transfected fish to provide a transgenic fish having the characteristics of the gene sequence.

The invention provides a method for determining the genomic sex of various salmonids (family salmonidae). In particular, the invention provides the nucleic acid sequence of a pseudogene, designated GH-.PSI., which is linked to a sex determining locus on the Y chromosome, and may be used as a marker for determination of the sex of the fish.
