Provided is a method for labeling a nucleic acid, including providing a target nucleic acid to be labeled; providing a 5'-end glycosylase to react with the target nucleic acid to create an intermediate nucleic acid having an abasic site at a 5′-end; and providing an aldehyde-reactive compound carrying a detectable label for coupling with the intermediate nucleic acid to form a labeled nucleic acid with the detectable label attached at 5′-end.
