An operation method of a multiplex slide plate device is provided. First, the multiplex slide plate device is assembled, including a slide plate, a sacrificial layer and a housing. The slide plate has reaction vessels, and the sacrificial layer has a microfluidic channel composed of an injection channel, a main channel and a distal channel. A sample solution is injected to the injection channel, such that the sample solution flows from the injection channel through the main channel to the distal channel, wherein the sample solution loads into the reaction vessels. Afterwards, an oil is injected to the injection channel, such that the oil flows from the injection channel through the main channel to the distal channel, wherein the oil removes the sample solution not loaded into the reaction vessels. Next, the sacrificial layer is heated to melt, and the melted sacrificial layer is mixed with the oil.

Operation method of multiplex slide plate device

A multiplex slide plate device is provided, including a slide plate, a sacrificial layer, and a housing. The slide plate has reaction vessels arranged in an array. The sacrificial layer has a microfluidic channel, which has an injection channel, a main channel, and a distal channel connected to each other. The housing is used to accommodate the slide plate and the sacrificial layer, and is composed of a cover and a tray, and the cover has an injection hole, an exhaust hole, and a storage tank. A sample solution and an oil are injected from the injection hole into the injection channel, wherein the sample solution is pushed by the oil. The sample solution loads into each of the reaction vessels while flowing through the main channel. Excess waste liquid flows from the exhaust hole into the storage tank, and is covered by the oil and cannot reflow.

Multiplex slide plate device having storage tank

A multiplex slide plate device and an operation method thereof are provided. The multiplex slide plate device includes a slide plate and a sacrificial layer. The slide plate has reaction vessels arranged in an array, an injection hole and an exhaust hole, wherein each of the reaction vessels has an opening portion and a bottom portion. The sacrificial layer has a microfluidic channel, wherein the microfluidic channel has an injection channel, a main channel and a distal channel connected to each other. The sacrificial layer is assembled to the slide plate, wherein the main channel faces the opening portion. A sample solution is injected into the injection channel, such that the sample solution flows from the injection channel through the main channel to the distal channel, wherein the sample solution loads into each of the reaction vessels while flowing through the main channel.

Multiplex slide plate device and operation method thereof

A measuring method for amplicon length is provided. A qPCR master mix, a forward primer, a reverse primer, a hybridization probe, a double-stranded DNA binding dye, and nucleic acid samples are added into reaction wells for qPCR reaction, and the fluorescence intensity of each of a hybridization probe and a double-stranded DNA binding dye varying with cycle number is respectively measured. Afterwards, amplicon length is obtained by applying a calculating method.

The present invention relates to a multiplex slide plate for various types of assays. The slide plate may be pre-filled with special-formulated reagents in different reaction zones, and the reactions carry out independently in the reaction zones filled with special-formulated reagent.
