Clinical information, molecular information and/or computer-generated morphometric information is used in a predictive model for predicting the occurrence of a medical condition. In an embodiment, a model predicts risk of prostate cancer progression in a patient, where the model is based on features including one or more (e.g., all) of preoperative PSA, dominant Gleason Grade, Gleason Score, at least one of a measurement of expression of AR in epithelial and stromal nuclei and a measurement of expression of Ki67-positive epithelial nuclei, a morphometric measurement of average edge length in the minimum spanning tree (MST) of epithelial nuclei, and a morphometric measurement of area of non-lumen associated epithelial cells relative to total tumor area. In some embodiments, the morphometric information is based on image analysis of tissue subject to multiplex immunofluorescence and may include characteristic(s) of a minimum spanning tree (MST) and/or a fractal dimension observed in the images.

Method and system for determining the risk of occurrence of prostate cancer

Clinical information, molecular information and/or computer-generated morphometric information is used in a predictive model for predicting the occurrence of a medical condition. In an embodiment, a model predicts whether a patient is likely to have a favorable pathological stage of prostate cancer, where the model is based on features including one or more (e.g., all) of preoperative PSA, Gleason Score, a measurement of expression of androgen receptor (AR) in epithelial and stromal nuclei and/or a measurement of expression of Ki67-positive epithelial nuclei, a morphometric measurement of a ratio of area of epithelial nuclei outside gland units to area of epithelial nuclei within gland units, and a morphometric measurement of area of epithelial nuclei distributed away from gland units. In some embodiments, quantitative measurements of protein expression in cell lines are utilized to objectively assess assay (e.g., multiplex immunofluorescence (IF)) performance and/or to normalize features for use within a predictive model.

System for evaluating a pathological stage of prostate cancer

This invention provides a method for determining the aggressiveness of a prostate carcinoma comprising: (a) obtaining a sample of the prostate carcinoma; and (b) detecting the presence of p27 protein in the prostate carcinoma, the absence of p27 indicating that the prostate carcinoma is aggressive. This invention also provides a method for diagnosing a beign prostate hyperplasia comprising: (a) obtaining an appropriate sample of the hyperplasia; and (b) detecting the presence of the p27 RNA, a decrease of the p27 RNA indicating that the hyperplasia is beign. This invention provides various uses of p27 in prostate cancer. Finally, this invention also provides different marker for prostate cancer.

Markers for prostate cancer

The present invention provides a method for creating a frozen tissue array. An oil in a liquid form is added into a recipient block containing frozen tissue cores where the oil has a freezing point lower than the freezing point of the tissue cores; and the recipient block containing the oil is cooled to a temperature about equal to or below the freezing point of the oil; the subsequently frozen oil locks the frozen cores in the recipient block. The oil may also be added to the recipient block prior to inserting the frozen tissue cores. The recipient block may be formed using a cryoarray device comprising a mold plate, an ejector plate, mold alignment pins, ejector pins, and cryoarray pins. Such method may be used for preparing frozen sections with multiple tissue specimens for assays such as in-situ hybridization and immunohistochemistry.

This invention provides a method of diagnosing a predisposition to cancer in a subject comprising: (a) obtaining a nucleic acid sample from the subject; and; (b) determining whether one or more of the subject's Ku70 alleles or regulatory regions to those alleles are deleted or different from the wild type so as to reduce or eliminate the subject's expression of polypeptide having tumor suppressor activity. This invention also provides a method of assessing the severity of cancer in a subject comprising: (a) obtaining a nucleic acid sample from the subject; and (b) determining whether one or more of the subject's Ku70 alleles or regulatory regions to those alleles are deleted or different from the wild type so as to reduce or eliminate the subject's expression of polypeptide having tumor suppressor activity. This invention also provides a method of assessing the severity of cancer in a subject comprising: determining the subcellular localization of Ku70 in the subject, wherein an abnormal subcellular localization of Ku70 indicates a predisposition to cancer.

Uses of Ku70

The present invention provides a method for creating a cryoarray of frozen tissue cores using a cryoarray system. Such system comprises a tissue mold, an embedding medium and a cryoarray device. The cryoarray device comprises a mold plate, an ejector plate, mold alignment pins, ejector pins, and cryoarray pins. Such method/system may be used for preparing frozen sections with multiple tissue specimens for assays such as in-situ hybridization and immunohistochemistry.

The subject invention describes a method of determining the secretor status of an individual which comprises obtaining a sample of a biological fluid from the individual and determining whether the sample includes the Lewis.sup.a or Lewis.sup.b antigens, the presence of the Lewis.sup.a antigen in the sample indicating that the individual is a nonsecretor, the presence of the Lewis.sup.b antigen in the sample indicating that the individual is a secretor, and the presence of neither antigen indicating the secretor status of the individual is inconclusive. The invention also provides a method of further determining the secretor status of an individual of having an inconclusive secretor status which comprises determining whether the biological fluid sample from the individual includes A, B or precursor type 1 chain antigens, the presence of any such antigens in the sample indicating that the individual is a secretor, the lack of any such antigens in the sample indicating that the individual is a nonsecretor. The invention provides a method of determining whether a human female subject is susceptible to urogenital infection which comprises determining whether the subject is a secretor according to the hereinabove-described methods, a secretor being susceptible to such an infection. The present application also provides a method of disstinguishing urothelial carcinoma from normal tissue, and identifying human germ cell tumor as seminoma or nonseminoma.
