The present invention providesendoglucanase I variants exhibiting improved cellulase and/or xylanase activity, in particular at 30&#xb0; C. and/or pH 6. These variants are particularly useful in industrial processes comprising simultaneous degradation of cellulosic biomass into monomeric sugars and fermentation of said sugars, for example to produce ethanol.

The invention relates to a recombinant DNA molecule encoding a polypeptide having phytase activity and increased temperature stability and increased proteolytic stability of the enzyme activity. The DNA sequence has been obtained by variation of the mature wild-typephytase sequence with defined amino acid positions being modified in comparison to the wild-type sequence or with the sequences having N- and/or C-terminal extensions, respectively. The invention further relates to a method for expressing the recombinant phytase as well as its use in the food and animal feed technologies.

Polypeptides having phytase activity and the encoding nucleic acids

The present invention provides the field of enhancing proteins and in particular to that of proteins enhanced by molecular change. It provides a variant of a phytase that is termed enhanced in that is has better thermostability and/or activity than the original phytase. The invention also provides a nucleic acid coding for said variant, a cassette or an expression vector containing said variant, a host cell expressing said variant, a composition comprising said variant and uses thereof, principally in the preparation of food additives and animal feed.

Enhanced phytase variants

The invention relates to a method for recovering of oil from plant seeds characterized in that a) an aqueous solution containing one or more cellulolytic and/or lipolytic and/or pectinolytic and/or proteolytic enzyme(s) and/or phytase is sprayed onto the seed, b) the thus obtained seed is directly supplied to a one-stage or multistage pressing in a way known per se, optionally coupled to an extraction, and c) the oil is recovered in a way known per se and optionally further processed, and the use of the method, particularly in the production of edible oil or biodiesel.

The present invention relates to a microbial expression system for the production of polypeptides based on the use of extrachromosomal DNA, whereby no antibiotic marker genes for the selection of the host cell but DNA sequences that encode glycerine-3-phosphate dehydrogenase are used, and, thus, the production of the desired polypeptide, e.g., xylanase, does not need the addition of antibiotics. The present invention features a recombinant DNA molecule which, upon expression in a prokaryotic or eukaryotic host cell, encodes a polypeptide having phytase activity. In preferred embodiments, the recombinant DNA molecule comprises a DNA sequence selected from DNA sequences which have been obtained by variations of the mature wild-typephytase sequence, wherein at least one amino acid at position 200 or position 207 is mutated as compared to the wild-type sequence, where the recombinant DNA molecule is, upon expression in a suitable host cell, associated with an increased activity of the thus encoded protein in the culture supernatant.
