An opioid independent surgical anesthetic composition includes an injectable dosage form of a hydrogel having a plurality of solid lipid matrix particles entrapped therein. The solid lipid matrix particles include a lipophilic local anesthetic drug and a lipid glyceride (e.g., saturated triglyceride or lipid blend of various lipid glycerides). Methods for creating a long-acting local anesthetic product can include creating a bulk solid of a lipid matrix product by heating a lipid solvent above its melting point, dissolving a lipophilic local anesthetic drug therein, reducing a temperature of the resultant drug-lipid solution to below the melting point of the lipid solvent, and heat annealing the lipid matrix to remove or reduce presence of any unstable polymorphs in the lipid matrix. The methods can further include crushing the bulk solid of the lipid matrix product to form solid lipid matrix particles and entrapping the solid lipid matrix particles within a hydrogel.

Opioid independent surgical anesthetic

Sample preparation can include obtaining a sample in the form of a liquid mixture and forcing the liquid mixture through a cavitation chamber at an optimal pressure for separating pathogens from particles in the mixture without fragmenting at least 30% of the pathogens. Apparatuses used for sample preparation methods can include a fluid circuit, a cavitation chamber incorporated into the fluid circuit and having a channel with first, second, and third cross-sectional areas, the second cross-sectional area being downstream of and smaller than the first cross-sectional with respect to fluid flow through the fluid circuit and the third cross-sectional area being larger than and downstream of the second cross-sectional area with respect to fluid flow through the fluid circuit. The apparatus can include a pump in fluid communication with the cavitation chamber and a pressure sensor positioned upstream of the cavitation chamber.

Hydrodynamic cavitation

A tissue sample processing system and associated methods is disclosed and described. The tissue sample processing system () can include a microfluidic separating system (). The microfluidic separating system () can include a fluid channel to receive a carrier fluid () and a tissue sample (), and a plurality of outlets. Flow of the carrier fluid () and the tissue sample () in the fluid channel can facilitate segregation of materials in the tissue sample () based on size into a plurality of size fractions, such that each one of the plurality of outlets receives a different size fraction of the materials in the tissue sample. In addition, the sample processing system () can comprise a cryopreservation system () associated with at least one of the plurality of outlets to freeze the material in the tissue sample () associated with the at least one of the plurality of outlets.

Tissue sample processing system and associated methods

Implementations of the present disclosure relate to apparatuses, systems, and methods for anastomosing vascular systems in medical procedures. A pair of similar or identical coupling devices may be disposed at the ends of two vessels, either natural or synthetic. The coupling devices may be capable of hermaphroditically connecting to one another to provide a simple and secure connection which promotes growth of the vessels between the bio-absorbable coupling devices.

A method for separating and enriching sperm from a tissue sample comprises: obtaining a microfluidic separating system having an inlet end and an outlet end, and a membrane filter (e.g., hollow fiber membrane filter) fluidly connected to the outlet end; separating the tissue sample via the microfluidic separating system into a debris fluid volume and a sperm fluid volume; and enriching the sperm fluid volume by removing excess media via the membrane filter. A two-stage tissue sample separation system comprising: a microchannel structure defining a separation fluid channel to form a separation stage; an inlet end of the microchannel structure; an outlet end of the microchannel structure; and a membrane filter fluidly connected to the outlet end for removal of at least a portion of excess media in the tissue sample.

Microfluidic system for sperm separation and enrichment from various types of sperm samples

A device for rapid non-destructive genetic material collection can include a multi-reservoir array () and a movement mechanism. The multi-reservoir array () can include multiple reservoirs (). A plurality of the multiple reservoirs () can include an abrasive surface () capable of retaining a source of genetic material in a liquid carrier. The abrasive surface () has a roughness. The movement mechanism can be operable to move the multi-reservoir array () in an oscillating motion sufficient to create relative movement between the abrasive surface () and the source of the genetic material in order to remove a portion of genetic material from the source of the genetic material without destroying the source of the genetic material or the portion of the genetic material that is removed.

Rapid non-destructive genetic material collection

A nerve repair conduit configured to be secured on first and second portions of a selected nerve. The nerve repair conduit includes a polymeric body having a proximal end, a distal end, an exterior surface and an interior surface defining an interior lumen. In addition, the nerve conduit includes at least one drug reservoir to hold agent(s) that may facilitate nerve regeneration. The drugs diffuse from the drug reservoir(s) into the nerve repair conduit through an outlet (such as a hole or a semipermeable membrane) in proximity to the first and second portions of a selected nerve. The nerve repair conduit may be configured to deliver the agent(s) at a rate having substantially zero-order kinetics and/or at a constant rate over a selected period of time.

Methods and devices for connecting nerves

A nerve repair conduit configured to be secured on first and second portions of a selected nerve. The nerve repair conduit includes a polymeric body having a proximal end, a distal end, an exterior surface and an interior surface defining an interior lumen. In addition, the nerve conduit includes at least one drug reservoir to hold agent(s) that may, for example, facilitate nerve regeneration. The drugs diffuse from the drug reservoir(s) into the nerve repair conduit through an outlet (e.g., a semipermeable membrane) in proximity to the first and second portions of a selected nerve. The nerve repair conduit may be configured to deliver the agent(s) at a rate having substantially zero-order kinetics and/or at a constant rate over a selected period of time (e.g., at least 1 week).

Devices, systems, and methods for delivery of an active agent from the lens capsule to a posterior segment of the eye of a subject can include an intraocular active agent delivery device including an active agent dispersed within a biodegradable active agent matrix. The active agent includes dexamethasone and the delivery device is adapted to fit within a lens capsule or ciliary sulcus of an eye. The delivery device can be inserted into the lens capsule or ciliary sulcus of an eye during cataract surgery or for treatment of uveitis.

Intraocular drug delivery device and associated methods

A microfluidic device can include a temperature regulator positioned to generate a temperature gradient across a thermally controlled portion of the microfluidic device from a high temperature region to a low temperature region. Additionally, the microfluidic device can include a bi-directional microfluidic serpentine pathway having a first terminus and a second terminus. The bi-directional microfluidic serpentine pathway can be oriented along a longitudinal direction transverse to the temperature gradient. Further, the bi-directional microfluidic serpentine pathway can include a plurality of oscillation segments fluidly coupling the first terminus to the second terminus and forming a flow path adapted to oscillate a fluid between the high temperature region and the low temperature region. The bi-directional microfluidic serpentine pathway can have a uniform cross section to facilitate bi-directional flow.

The present disclosure provides apparatuses, systems, and methods involving a spotter for depositing a substance on a submerged surface. The spotter comprises an outlet cavity defined at least in part by a spotting orifice, a first opening, and a second opening. The spotter also comprises a first conduit fluidly coupled to the first opening and a second conduit fluidly coupled to the second opening. The spotter is adapted so that fluid flowing through the first conduit and the second conduit is communicated among the first opening, the second opening, and a submerged deposition surface when the sealing orifice is sealed against the submerged deposition surface to form a deposition spot on the submerged deposition surface. The submerged deposition surface is within a liquid such that the liquid covers the deposition spot upon removal of the orifice from the deposition surface.

Method and device for depositing a substance on a submerged surface

The present disclosure provides apparatuses, systems, and methods involving a spotter apparatus for depositing a substance from a carrier fluid onto a deposition surface in an ordered array. The spotter apparatus includes a loading surface, including a first well and a second well, and a different outlet surface, including a first opening and a second opening, where a first microconduit fluidly couples the first well with the first opening and a second microconduit fluidly couples the second well with the second opening. An overlay is sealed to the outlet surface and penetrated by a deposition channel that is situated to communicate carrier fluid among the first opening, the second opening, and the deposition surface when the overlay is pressed against the deposition surface.
