Electrophysiologic biomarkers for prognostication of neurological outcome are described herein. An inverse correlation was found between timing of a cortical spreading depolarization (SD) wave and neurological outcome as tested at 24 hours post-CPR. Additionally, a minor image of this SD was identified as a 'repolarization (RP) wave.' Quantifying features of SD and RP during cardiac arrest and cardiopulmonary resuscitation (CPR) provide important metrics for diagnosis and prognosis of neurological injury from hypoxia-ischemia and can serve as an early prognostication tool for predicting outcome at subsequent days after successful CPR. This discovery may also allow for novel therapeutic interventions to improve neurological recovery after hypoxia-ischemia insults.

Spreading depolarization and repolarization as biomarkers of neurological recovery after cardiac arrest

A multimodal optical imaging platform is used to obtain cerebral perfusion-metabolism mismatch metrics for rapid assessment of acute brain injury, ongoing (real-time) feedback to optimize cardiopulmonary resuscitation to improve neurological outcome, and rapid prognosis of recovery. Light of several wavelengths and types is delivered to the tissue, which is then absorbed and scattered by tissue components such as blood and cellular components. Some of this light scatters back to the surface, where it is captured by a detector. The resulting data are processed to obtain blood flow and oxygenation parameters, as well as tissue scattering. These parameters are then combined to calculate metabolism and flow-metabolism coupling/decoupling metrics, which are used to determine ischemic damage, ongoing need for optimal blood flow and oxygenation, and to predict cerebral recovery in patients with acute brain injury during and immediately after cardiac arrest, stroke, traumatic brain injury, etc.

Real-time methods to enable precision-guided CPR to improve neurological outcome and predict brain damage after ischemic injury and reperfusion

An in vivo optical biopsy applicator of the vaginal wall for treatment planning, monitoring, and imaging guided therapy is described herein. The applicator may include an imaging probe operatively coupled to a laser ablation device. The applicator allows for non-invasive optical tissue monitoring in order to define pre- and post menopausal parameters, pre- and post-treatment microscopic changes, and offers an objective scientific tool in order to compare currently available medical, non-medicated, and energy-based treatment protocols.

Optical biopsy applicators for treatment planning, monitoring, and image-guided therapy

In accordance with various embodiments herein, disclosed herein is an apparatus comprising an imaging probe-based device operably linked to an automated tracking system. Furthermore, in various embodiments, the apparatus may be used to track the position and movement of a handheld imaging probe, including with respect to a subject's 3D surface profile. In one embodiment, a device may be used to track the position and movement of a handheld imaging probe with respect to a subject's 3D surface profile, while simultaneously acquiring optical data.

Motion tracking apparatus and method

A quantitative, non-invasive optical spectroscopy technique for measuring dynamic changes in adipose tissue structure and metabolism in vivo. The technique requires multiple wavelengths of light in the near-infrared (650-1000 nm). Using these wavelengths of light, adipose tissue is illuminated and light that returns to a photodetector is analyzed. From the return signals, the absorption and reduced scattering coefficients (&#x3bc;and &#x3bc;') at each wavelength are calculated. The obtained &#x3bc;and &#x3bc;&#x2032; values allow for quantification of biomarkers and indices which allow for measurement of fat composition and metabolism. The concentration of oxy- and deoxy-hemoglobin, the fractional water and lipid content, and information about the size distribution of light scatterers in the adipose tissue are also determined. A detailed and quantitative understanding of fat composition and metabolism is thereby provided which describes the effectiveness of interventions to improve the health of a patient.

Predicting weight loss and fat metabolism using optical signal changes in fat

Systems and methods are disclosed for determining physiological information in a subject. The system includes: a light source positionable along a first location outside of the subject; a photo-sensitive detector positionable along a second location outside of the subject and configured to detect scattered light and generate a signal; a processor having a program and a memory, wherein the processor is operably coupled to the detector and configured to receive and store the signals generated over a period of time; wherein the processor is programmed to derive contrast metrics from the stored signals, calculate a waveform from the contrast metrics, decompose the waveform into basis functions and respective amplitudes, and compare the basis function amplitudes to determine the physiological information.

Non-invasive hemodynamic assessment via interrogation of biological tissue using a coherent light source

A low cost portable high speed quantitative system for diffuse optical spectroscopic imaging of human tissue. The hybrid system (CWFD) can measure absolute optical properties from 660 nm to 980 nm and recover all tissue chromophore concentrations. The standalone FD module can be utilized to measure scattering at every measurement and recover deoxygenated and oxygenated hemoglobin concentrations. The CW module can operate concurrently with the FD module to also measure water and lipid. The high temporal resolution and large signal-to-noise ratio of the CWFD system may be used to explore tissue oximetry, vascular occlusion, and paced breathing models to measure and analyze tissue hemodynamics response to changes in blood flow. Continuous monitoring of vasculature response to various modified blood perfusion conditions can provide information about local tissue metabolism and physiological state (dysfunction).

Hand-held optical scanner for real-time imaging of body composition and metabolism

A multiphoton microscope based on two-photon excited fluorescence and second-harmonic generation that images FOVs of about 0.8 mm(without stitching adjacent FOVs) at speeds of 10 frames/second (800&#xd7;800 pixels) with lateral and axial resolutions of 0.5 &#x3bc;m and 2.5 &#x3bc;m, respectively. The scan head of the instrument includes a fast galvanometric scanner, relay optics, a beam expander and a high NA objective lens. The system is based on a 25&#xd7;, 1.05 NA water immersion lens, which features a long working distance of 1 mm. A proper tailoring of the beam expander, which consists of the scan and tube lens elements, enables scaling of the FOV. The system and method also include a flat wavefront of the beam, minimum field curvature, and suppressed spherical aberrations. All aberrations in focus are below the Marechal criterion of 0.07&#x3bb; rms for diffraction-limited performance.

Imaging platform based on nonlinear optical microscopy for rapid scanning large areas of tissue

The present invention relates to methods and devices that may be used to extract spatial frequency information. In one embodiment, the invention provides an instrument configured to extract data using a two-dimensional (2D) Hilbert transform technique, and providing spatial frequency information from a sample. In another embodiment, the invention provides a spatial frequency domain imaging (SFDI) device adapted for demodulation using two or less imaging frames.

Method for extraction of spatial frequency information for quantitative tissue imaging

A diffuse optical spectroscopic imaging (DOSI) apparatus for tissue spectroscopy measures absorption and scattering properties of tissue using multi-frequency frequency domain photon migration in a modular or networkable platform to provide full broadband information content. The apparatus includes: a broadband signal generator; a driver having an input coupled to the signal generator; a light source coupled to the driver, the light source for exposing the tissue to broadband modulated light at a plurality of wavelengths; an optical detector for receiving returned light from the tissue; an amplitude detection circuit communicated to the optical detector; a phase detection circuit communicated to the optical detector; and a plurality of filters and amplifiers, wherein the optical detector, amplitude detection circuit and phase detection circuit are interconnected with each other by corresponding ones of the plurality of filters and amplifiers to isolate signals and increase signal-to-noise ratio.

Portable broadband diffuse optical spectroscopic imaging (DOSI) device for noninvasive tissue characterization

A fiber-delivered probe suitable for CARS imaging of thick tissues is practical. The disclosed design is based on two advances. First, a major problem in CARS probe design is the presence of a very strong anti-Stokes component in silica delivery fibers generated through a FWM process. Without proper spectral filtering, this component affects the CARS image from the tissue sample. The illustrated embodiments of the invention efficiently suppress this spurious anti-Stokes component through the use of a separate fiber for excitation delivery and for signal detection, which allows the incorporation of dichroic optics for anti-Stokes rejection. Second, the detection of backscattered CARS radiation from the sample is optimized by using a large core multi mode fiber in the detection channel. This scheme produces high quality CARS images free of detector aperture effects. Miniaturization of this fiber-delivered probe results in a practical handheld probe for clinical CARS imaging.

System and method for efficient coherence anti-stokes raman scattering endoscopic and intravascular imaging and multimodal imaging

A single modulated imaging platform is employed. Method and apparatus for performing qualitative and quantitative analysis of produce (fruit, vegetables) using spatially structured illumination

An improvement in a method for quantitative modulated imaging to perform depth sectioned reflectance or transmission imaging in a turbid medium, such as human or animal tissue is directed to the steps of encoding periodic pattern of illumination preferably with a fluorescent excitation wavelength when exposing a turbid medium to the periodic pattern to provide depth-resolved discrimination of structures within the turbid medium; and reconstructing a non-contact three dimensional image of the structure within a turbid medium. As a result, wide field imaging, separation of the average background optical properties from the heterogeneity components from a single image, separation of superficial features from deep features based on selection of spatial frequency of illumination, or qualitative and quantitative structure, function and composition information is extracted from spatially encoded data.
