The present invention relates to antibodies and antigen-binding portions thereof that specifically bind to insulin-like growth factor I receptor (IGF-IR), which is preferably human IGF-IR. The invention also relates to human anti-IGF-IR antibodies, including chimeric, bispecific, derivatized, single chain antibodies or portions of fusion proteins. The invention also relates to isolated heavy and light chain immunoglobulin molecules derived from anti-IGF-IR antibodies and nucleic acid molecules encoding such molecules. The present invention also relates to methods of making anti-IGF-IR antibodies, pharmaceutical compositions comprising these antibodies and methods of using the antibodies and compositions thereof for diagnosis and treatment. The invention also provides gene therapy methods using nucleic acid molecules encoding the heavy and/or light immunoglobulin molecules that comprise the human anti-IGF-IR antibodies. The invention also relates to gene therapy methods and transgenic animals comprising nucleic acid molecules of the present invention.

Antibodies to insulin-like growth factor I receptor

The present invention relates to a therapeutic method for the treatment of non-hematologic malignancies comprising administering anti-IGF-1R antibodies, particularly human anti-IGF-1R antibodies, to a patient, in conjunction with the administration of at least one other therapeutic agent. The invention further relates to pharmaceutical compositions comprising these antibodies and methods of using such compositions thereof for treatment.

Combination treatment for non-hematologic malignancies

The present invention relates to antibodies, which are directed to the human IGF-1 receptor (IGF-1R) and are to be administered for the treatment of cancer. The antibodies of the present invention have been altered to comprise antibodies with one or more selected germline framework amino acid residues which replace one or more corresponding somatically mutated residues in the variable region of the unaltered antibody. The modification results in the framework region mutations converted to germline. The modification results in a reduced propensity for the antibody to elicit an immune response (reduced immunogenicity) following administration to a human subject.
