Tissue dissociators configured to disrupt a biological tissue sample are provided. Aspects of the tissue dissociators may include a housing having a distal end and a proximal end, a cutting blade positioned at the distal end of the housing and a tissue actuator configured to be displaced along a longitudinal axis within the housing. Also provided are methods of using the tissue dissociators, as well as kits including the tissue dissociators.

Devices and methods for dissociating a biological tissue sample

Methods and devices for exchanging therapeutic agents, such as cells, from one liquid medium to another liquid medium are provided. Aspects of embodiments of the methods include transferring a therapeutic agent from a first medium, such as a freezing, storage or shipping buffer, into a second medium, such as a sterile physiologically compatible buffer. In certain aspects, the transfer of the therapeutic agent from a first medium to a second medium involves the use of acoustic-focusing, or acoustophoresis. Embodiments of the subject methods may facilitate the transfer of a therapeutic agent from a storage medium to an infusion medium, and in certain embodiments include administering the therapeutic agent contained in the infusion medium to the subject. Also provided by the present disclosure are devices for practicing the subject methods.

Fluid exchange methods and devices

Devices for sorting components (e.g., cells) contained in a liquid sample are provided. In certain aspects, the devices include a magnetic separation device and an acoustic concentrator device fluidically coupled to magnetic separation device. Aspects of the invention further include methods for sorting cells in a liquid sample, and systems, and kits for practicing the subject methods.

Devices and methods for manipulating components in a fluid sample

Systems for sterile separation of magnetically labeled sample components and methods for using the same are provided. Embodiments of the systems include a magnetic separation device and a pliant sample container, where a portion of the pliant sample container is operatively coupled under pressure to the magnetic separation device. Also provided are methods of using the systems, as well as pliant sample containers configured for use with the subject systems and methods.

Flow cytometric systems for sterile separation of magnetically labeled sample components

The present invention provides primers and probes to be used in a method of enhancing hybridization of a probe to a target nucleotide sequence when the target sequence is capable of forming intramolecular secondary structures that interfere with hybridization of the probe to the target sequence. In particular, the invention includes a primer for amplifying a target nucleotide sequence, wherein at least a portion of the target nucleotide sequence can form an intramolecular secondary structure. The primer of the invention includes a primer nucleotide sequence complementary to a portion of the target nucleotide sequence that does not form a secondary structure, and a blocking sequence substantially complementary to at least a portion of the secondary structure-forming region of the amplified target nucleotide sequence, wherein the blocking sequence hybridizes to a portion of the secondary structure-forming region of the amplified target nucleotide sequence and blocks the formation of the secondary structure.

Dual-purpose primers and probes for providing enhanced hybridization assays by disruption of secondary structure formation

The invention provides a set of highly orthogonal six-code universal sequences for use in bDNA singleplex and multiplex nucleic acid hybridization assays. The six-code orthogonal sequences do not cross-hybridize and thus, minimize or eliminate the 3-mer cross-hybridization inherent in the second and third generation bDNA assays. The highly orthogonal universal sequences may be used in singleplex or multiplex bDNA assays quantitatively and qualitatively to determine mRNA levels in a sample; to screen for and genotype targets, such as viruses, that are present in low volumes in a sample; to screen for and genotype SNPs; and to measure changes in the amount of a gene in a sample such as when gene amplifications or deletions occur. The highly orthogonal universal sequences may also be used as universal capture probes to selectively bind assay components in a way that facilitates their further analysis.

Highly orthogonal universal sequences for use in nucleic acid assays

Methods of determining the presence or amount of a target polynucleotide in a sample are provided. A sample that contains a target polynucleotide, a polynucleotide that may include nucleic acid analogs and that is complementary to a target nucleic acid sequence of the target polynucleotide, and a dye for which the rate of change in an optical property is different in the presence and absence of a target polynucleotide/polynucleotide hybrid are combined to produce a reaction mixture. The rate of change in an optical property of the dye in the reaction mixture is compared to a reference value characteristic of the rate of change in the optical property of the dye in a similar reaction mixture containing a known amount of a polynucleotide/polynucleotide hybrid to determine a relative rate of change in the optical property. The relative rate of change in the optical property of dye in the reaction mixture is correlated with the presence or amount of the specified target polynucleotide in the sample.

Methods and compositions for detecting polynucleotides

The present invention relates to the methods for assaying an analyte comprising a nucleic acid analog binding substrate in a sample, comprising reacting a catalytic complex comprising a nucleic acid analog, a nucleic acid analog specific binding substrate and a light reactive dye with a light stimulus, and detecting the presence or absence or amount of a reaction product of the catalytic complex and light stimulus. Methods and compositions for rapid light-activated isolation and detection of analytes

A bucket holding apparatus includes first and second carriages with pivotally attached inner edges arranged in such a manner that the first and second carriages freely rotate upwardly from a horizontal plane defined on a support surface. The bucket holding apparatus further includes first and second arcuate walls directly conjoined to the first and second carriages and oppositely situated from the inner edges such that the first and second arcuate walls define a circular cavity within a combined perimeter thereof. Methods are provided for reducing background signals encountered in nucleic acid hybridization assays and other assays that involve hybridization of a labeled oligomer to its complement. The method is premised on the significant reduction of signal generation that occurs when a quenchable dye-labeled oligomer forms a hybrid complex.
